Aim: To further investigate effect of flavonoids extracted from Lysimachiaclethroides Duby (ZE4) on hepatom and uterine cervix cancer and its mechanism.Methods: MTT assay, 3H-TdR and colony-forming units assay were used to selectsensitive tumor cell lines and to determine inhibitory effects of ZE4 after various inhibitorswere added. Then we investigated the inhibitory effects of ZE4 on the transplantable tumormodels of H22 and U14. The anti-migration effects of ZE4 were observed by scratch test andadhesion test. The artificial lung metastasis models of mice were established to see theanti-invasive ability of ZE4 in vivo. We used AO/EB and Hoechst fluorescent staininganalysis and cell immunofluorescence study to further investigate the anti-tumormechanism of ZE4. Results: ZE4 could significantly inhibit proliferation and metastasison hepatoma and uterine cervix cancer in vivo and in vitro. In mechanism research, wefound that ZE4 could up-regulate the expression Fas/FasL, TRAIL/DR5 and caspase-3gene and down-regulate the expression Bcl-2 and VEGF. It could induce apoptosis inSMMC-7721 cells. Conclusion: ZE4 could inhibit proliferation and metastasis onhepatoma and uterine cervix cancer, its mechanism was associated with the regulation ofseveral functional gene and protein.
