Liver cancer is one of the common malignant tumors. The foundation which themalignant tumor occurs is the non-balance match and the shift proliferation which thecell multiplication and apoptosis. The cell apoptosis is close with the liver canceroccurrence and development. The nuclear glucoside analog is one kind importantanti-cancer medicament.In this thesis, we have reviewed the recent advances in studies on the relationshipof liver cancer occurrence with cell apoptosis, the modulation and examinationtechnology of the cell apoptosis, the nuclear glucoside analog structure and thepharmacology, and the synthetic method research progress.Base the research of natural nucleic acid metabolize pathways in organism, takethe natural adenine nucleotide as the reference sample, two kind of novel adenineglucoside analog were synthesized from the adenine as raw material by the principle ofactivity superposition and electron equall-arrange. The nature nuclear glucoside wasembellished by introducing the sulfur and phosphorus element.The total compounds structure were determined by 13CNMR, 1HNMR, C-Hcosy,IR, MS and X-ray diffraction technology. The adenine glucoside analog containe thesulfur element to carry on the thermal analysis, has induced this kind of compoundthermal decomposition rule; the mechanism which the phosphorus class adenine glucoside analog induced liver cancer BEL-7402 cell apoptosis was studied by theMTT method, the DNA gel electrophoresis, the protein electrophoresis, the fingerprintpeptide spectrum, flow cytometry, and confirmed its pharmacology mechanism and theway initially.The contents and the primary contributions of the reseach are summarized asfollows:1. Take the adenine as the initial structure ,and choose the xylose, glucose,galactose and lactose as the synthesis factor and introduce the sulfur element. TheThio-hydrazide compounds contain deaced-saccharide group were synthesized and hascarried on the exploration to its synthesis condition.2 Take the adenine as the initial structure, The Phosphodiester adenine nucleotidecompounds were synthesized by the classic Arbuzov reaction and has carried on theexploration to its synthesis condition.3. The total compounds structure were determined by 13CNMR, 1HNMR, C-Hcosy,IR, MS and X-ray diffraction technology. The adenine glucoside analog containe thesulfur element to carry on the thermal analysis (TG, DSC), has induced this kind ofcompound thermal decomposition ruleThe mechanism which the phosphorus class adenine glucoside analog inducedliver cancer BEL-7402 cell apoptosis was studied in some different level include cell,cell organ, gene, protein and the dynamic cell cycle by the MTT method, the DNA gelelectrophoresis, the protein electrophoresis, the fingerprint peptide spectrum, flowcytometry, and confirmed its pharmacology mechanism is not to rely on bcl-2 butthrough the Caspase way.
