Objective: To research the effects of blood serum containing Decoction of TaoHong SiWuTang(THSWT) on vascular smooth muscle cells(VSMC) migration and the expression of integrinβ3, matrix metalloproteinase9(MMP9) and focal adhesion kinase(FAK)/P38 MAPK signal,and to investigate the mechanisms about it.Methods: Blood serum containing Decoction of THSWT and normal blood serum from rats were Prepared. The vascular smooth muscle cells were isolated from common carotid artery of rats, then were cultured in vitro, transfered of culture and identified by immunocytochemistry. Vascular smooth muscle cells were disposed to be cultured with normal blood serum(group normal serum) and blood serum containing Decoction of THSWT (group drug serum). Cell migration activity of each group was assayed by modified micro-Boyden chamber. The expression of integrinβ3, MMP9 and FAK of each group were detected by immunocytochemistry staining. Western blot was used for determining variance about the expression of P38MAPK of each group.Results: Cell migration existed in normal control group. After intervention with blood serum containing THSWT, the migrating cell population diminish and the migration lenth decurtated( P<0.01). Comparing with normal control group, after intervention with blood serum containing Decoction of THSWT, the expression of integrinβ3, MMP9, FAK and P38MAPK all decreased( P<0.01).Conclusion: blood serum containing THSWT could restrain vascular smooth muscle cells migration and could negatively influence the expression of integrinβ3, MMP9 and FAK/P38 MAPK signal as well, the influence play a role in the depressant effect on vascular smooth muscle cells migration or proliferation.
