| OBJECTIVE:To study the effect of direct bone morphogenetic protein 4 (BMP-4) gene therapy mediated by non-replicational adenovirus in bone formationMETHODS:Thirty Sprague-Dawley rats were used in this study.Bone defect with size of 0.8cm×0.5cm were made at the bilateral tibia of 30 male Sprague-Dawley rats by self-control method ,the right tibial bones were experimental sides and the left ones were as blank controls. the non-replicational adenovirus of human bone morphogenetic protein4 and glutin sponge were implanted into bone defect in experiment groups ,only glutin sponge was implanted in control groups.The X-ray, histological examination, immunoh- istochemical staining ,and transmission electron microscope were made 4, 6, and 8 weeks after the operation.RESULTS: All the 30 rats were involved in the analysis of results.①The result of histological and radiographical examinations showed that the defect of bone has been repaired both on right and left sides at 8 weeks,but the efficacies of the speed ,effect of the bone formation and the volume of new bones on the right were superior to those on the left.②experiment group gained better effect in the volume of new bones, and the expression of BMP-4 than those of control groups.CONCLUSION: The expression of target gene was indentified by the animal experiment and target protein has biological activity. direct BMP-4 gene therapy is easy to perform and has very strong osteoinduction ability. It is a good method for repairing the bone defect.. |
