【Objectives】1.To study the expression of in the different glucose concentration with the time changing.2.To study the effect of different concentration of Hepatocyte Growth Factor on the Integrin-linked kinase in protein and mRNA level by Mesangial Cells in high concentration (25 mmol/L) of glucose.3.To study the effect of different concentration of HGF on the Fibronectin'secretary by Mesangial Cells.4.To study latent therapeutic action and possible mechanism of HGF on the glomeruler sclerosis.【Methods】The difference of ILK's experession on mRNA level in different concentration of glucose and HGF are detected by means of revers transcription-polymerase chain reaction,and by means of immunohistochemistry to detect their difference in protein level;the effection of HGF to the FN's secretary in high glucose are detected by means of enzyme linked innunosorbent assay(ELISA).【Result】 1)By RT-PCR method we found that under the glucose concentration of 5.6 mmol/L and 11mmol/L with the increase of time the expression of ILK in mRNA level have no difference (p>0.05)。When the glucose concentration reach 25 mmol/L with the increase of time the ILK's expression in mRNA level increased and at 36 hours reach the top,although go on increasing the reaction time the ILK's expression will not increased longer.With the immunohistochemistry method we get the same conclusion: under the glucose concentration of 5.6 mmol/L and 11mmol/L with the increase of time the ILK'protein content have no change,the colours in endochylema have no difference;Under the 25mmol/L glucose dose when the reaction time get 36 hours the colours in endochylema become deeper distinctly.2)When the HGF'concentration get 20 ng/ml it will obviously inhibite the ILK'expression in mRNA level under the high glucose concention(25mmol/L) (P<0.05)。With the immunohistochemistry method when the HGF'concentration get 20 ng/ml the colours in endochylema become weaker distinctly.3)When the HGF'concentration get 20 ng/ml it will obviously inhibite the excretion of FN in Mesangial cell (P<0.05)4)Correlation analysis revealed that under the action of HGF the decreasion of the ILK'expression correlate with the secretion of FN in the mesangial cell.【Conclusion】1.When the glucose concention get 25 mmol/L it will obviously inhibite the ILK's expression,and at 36 hours get the top expression,increase the action time will not increase the ILK'expression any more.2.When the HGF get 20ng/ml it will obviously inhibite the ILK'expression in high glucose concentration(25mmol/L)3. When the HGF get 20ng/ml it will obviously inhibite secretion of FN in Mesangial cell.4.HGF have the potential therapy action to the diabetic nephropathy,and this action maybe lead by the ILK signal transduction,in order to inhibite the secreton of FN.
