The expression of TGF-β1 and Smad7 in gastric cancer tissue and corresponding normal mucosa tissueAIM :To detect the expression and disposition of TGF-β1 and TGF-β1 signal transduction negative regulator Smad7 in the gastric cancer tissue and the corresponding normal mucosal tissue, and to analyze the relations between the expression, disposition and the clinicopathologic characteristics, and to investigate the mechanisms of TGF-β1 and Smad7 in gastric cancer carcinogenesis and development.METHODS:40 cases of gastric cancer and normal tissues confirmed by histopathology were selected to detect and compare in the level of mRNA and protein by means of RT-PCR and frozen section immunohistochemistry staining technologyRESULTS:Thirty out of forty cases ( 75.0% )were positive for TGF-β1mRNA expression and 24 cases were positive for TGF-β1 staining. Strong staining of TGF-β1 was in the single carcinoma cell. There are significance between stage I group and stage II, stage III, stage IV group as far as TGF-β1 expression is concerned ( p<0.05 ). Also, significance were observed between invasion depth T1 group and invasion depth T2, T3, T4 group ( p<0.05 ). Nine out of eleven cases ( 81.8% ) were positive for Smad7 in group of T1. There are significant Smad7 expression between invasion depth T1 group and invasion depth T2, T3, T4 group ( p<0.05 ). TGF-β1 and Smad7 expression were prevalent in early gastric cancer. Frequently, strong TGF-β1 and Smad7 staining occurred in gastric signet-ring cell carcinoma. Smad7 and TGF-β1 expression were highly related ( r=0.79 ).. CONCLUSION:Blocking of TGF-β1 signal pathway by TGF-β1 signal transduction negative regulator s works in gastric cancer carcinogenesis and development. TGF-β1 and Smad7 appear to be molecule marker in the early gastric cancer diagnose, management and prognostic. Smad7 should be investigated as a potential target to restore TGF-β1 responsiveness and limit tumor growth. The expression and significance of TFF1 in the gastric cancer tissue and the corresponding normal mucoca tissueAIM: To check the expression and distribution in the gastric carcinoma tissue and the corresponding normal mucosa tissue and to investigate the probable roles the TFF1 plays in gastric cancer carcinogenesisMETHODS: 40 cases of exairesis gastric cancer and normal tissues confirmed by histopathology were selected to detect and compare in the level of mRNA and protein by means of RT-PCR and immunohistochemistry.RESULTS: TFF1 mRNA expression ratio 100.0% in the normal mucosa is higher than the expression in the cancer tissue47.5%, (p﹤0.05);TFF1 protein in the normal mucosa is more intense the expression in the cancer tissue, (p﹤0.05).TFF1 positive area was mainly focus on epithelia, glands and epithelial superficial mucous. The yellow pigmentation was in the cytoplasm, ,and the base of cell shows stronger pigmentation.CONCLUSION: Our results indicated that TFF1 expression in the normal mucosa is higher than the expression in the cancer tissue on the level of mRNA and protein, and The expression was in the cytoplasm, and the base of cell shows stronger pigmentation.
