| ã€objective】1. To investigate inhibitory effects of endostatin to angiogenesis of osteosarcoma. 2. To investigate inhibitory effects of endostatin to accretion and metastasis of osteosarcoma.ã€methods】1. The pBudCE4.1/ES was amplified, extracted and sequenced for appreciation. 2. Osteosarcoma cell line UMR106 was cultured in vitro. The endostatin was transfected in UMR106 through lipidosome and get the masculine cell clone with Zeocin. To observe the effect of endostatin to cell proliferation of UMR106 cell compared with control group and make the cell growth curve by MTT. 3. The endostatin excrete from the masculine cell was detected by ELISA. 4. To evaluate the activity of endostatin in vitro by supernatant culture with the vascular endothelial cell, and observe the reproductive activity of VEC. 5. To make the athymic mouse tumor model by injecting the masculine cell and UMR106 cell subcutaneouly, and observe the tumor growth, invasion, metastasize. 6. To make the athymic mouse tumor pulmonary metastasis model by injecting the masculine cell and UMR106 cell in vena caudalis,and observe the formation, amount and size of the tumor.ã€results】1. The pBudCE4.1/ES was amplified, extracted and accredited for the masculine recon. 2. The transfection is successful, and the endostatin was expressed by the UMR106 cell stable. The endostatin has the inhibitory effects to vascular endothelial cell proliferation. 3. Endostatin inhibits the accretion of hypotumor, but it does not effect the formation of tumor. 4. Endostatin inhibits the formation and accretion of pulmonary metastasis of UMR106 cell.ã€conclusions】Endostatin inhibits the accretion and metastasis of osteosarcoma, but it does not effect the formation of tumor. Endostatin can be used for osteosarcoma combined therapy. |
