The Experiment's Research Of Pre-emptive Analgesia By Intrathecal Injection With Buprenorphine

Objective:we use the model of rabbit't intrathecal injection to research the effects of pre-emptive analgesia with buprenorphine on rabbit's pian reaction and it's IL-2 and IL-6 producation.Methods:18 rabbits were randomly allocated into three groups(with 6 cases each):buprenorphine was given in GroupA as a experiment group, fentanyl was used in GroupB,and NS was used in Group C as a control group, after intrathecal injection 5minutes,inject of 5%formalin 0.5ml into the plantar surface of the right hind paw,Pain response was recorded with pain grade and two phase cumulative pain grade(the first phase is from 0 to 10minutes,and the second phase is from 10 to 45minutes)for a period of 45 minutes.We draw-off ear edge's venous blood 3ml at before analgesia and post-analgesia 2h,4h,8h,24 h and 48 h,finally,IL-2 and IL-6 were assayed by ELISA.Results:The pain response in the first phase(from 0 to 10minutes)was not inhibited in all three groups(P＞0.05);The pain response in the second phase(from 10 to 45minutes)was inhibited significantly in GroupA and GroupB compared with GroupC(P＜0.01).Compared with the values before analgesia,the level of IL-2 in Group C were decreased significantly at post-analgesia 48h(P＜0.05),and the lever was lower than that in the other groups at post-analgesia 48 h(P＜0.05);but no significantly difference between GroupA and GroupB(P＞0.05);at the others time the level of IL-2 was no significantly change(P＞0.05)and there were no significantly difference among groups(P＞0.05).In all three groups,the lever of IL-6 was no significant difference among groups(P＞0.05).Conclusions:Pre-emptive analgesia with buprenorphine and fentanyl can relieve the pain induced by formalin and restrain the decrease of IL-2,and it is efficient in maintaining celler immune function and the balance of celler interior environment in some sense.There was no difference with fentanyl in relieve the pain and suppress the immune suppression induced by the pain,but buprenorphine's effect was rapidly than fentanyl,and the two medicine both can be used in pre-emptive analgesia.
