Objective Through observing Nogo.B expressional changes in airways and lungs in asthma mice and its correlation to the activation of NF-kB to approach the role of Nogo-B in asthma and its probable mechanism in asthma pathological process.Methods Setting up asthma model,the changes of Nogo-B expression in airways and lungs were detected by immunohistochemistry,western blot and real time PCR.Its correlation to the activation of NF—kB was analyzed;the effects of dexamethasone on Nogo-B expression were observed. Results and conclusions Nogo-B immunologic-positive protein only localized in blood vessel endothelium,membrane and endochylema of alveolar epithelial cell;in asthma models,Nogo-B expression in airways and lungs significantly increased.Its expressional intensity was positively correlated to the activation of NF-kB.This indicates that Nogo—B participates in the pathological changes of asthma and probably plays a key role in asthma pathogenesis through activating NF—kB.Nogo—B expression in asthma mice significantly decreased after being treated with dexamethasone.This infers that glucocorticoid partly plays its anti-inflammatory role through inhibition of NF—kB activation by downregulation of Nogo expression.
