What kinds role of SOCS2 in the carcinogensis and development of breast carcinomas? Raccurt and Sutherland got the different result on researching the breast neoplasma. In order to identify the relationship between the SOCS2 and the breast neoplasma, we investgate the expression of SOCS2 in breast carcinoma tissues and the relation to clinical and biological parameters by immunohistochemistry. Real-time quantitative reverse transcription polymerase chain reaction (RQ-RT-PCR) technique was applied for detecting SOCS2 mRNA expression in the breast carcinoma. With screened concentration of estrogen (10-8 mol/L) and tamoxifen (10-6 mol/L) by MTT assay treated with MCF-7 72 hours, then detected the expressions of SOCS2 mRNA by Real-time quantitative reverse transcription polymerase chain reaction(RQ-RT-PCR) method and SOCS2 protein by immunohistochemistry. Determined quantitatively apoptosis cells of MCF-7 by TUNEL method. In conclusion, SOCS2 expression was inversely related to Ki-67, TNM classification, histological grades and lymphnode metastasis(P<0.05).The apoptotic index of SOCS2 positive group is higher than the other two groups. The SOCS2 expression was inverse to STAT5 in the breast tissue and cell. The role of SOCS2 maybe implement by STAT5 in the breast neoplasma. | | Keywords/Search Tags: | breast neoplasms, SOCS2, FQ-PCR, tissue microarray supressors of cytokine signaling, IHC, TUNEL, MTT, SOCS, STAT5, signal transducer | PDF Full Text Request | Related items |
