Objective: 42 familiar cancer cell lines were filtrated by CD133 antibody, to make sure whether there are cancer stem cells among these cancer cell lines. And the effects of apoptosis of HT29 and SW620 colon carcinoma cell line induced by oxaliplatin and 5-Fu were separately observed. the changes of quantity of CD133+ cell in HT29 and SW620 cells before and after chemotherapy were observed, and the effect of chemotherapy to CD133+ cell were detected. Methods: 42 logarithmic growth cancer cell lines were analyzed by flow cytometry after being marked by CD133 antibody.the proportion of CD133+ cell to each cell line were detected. HT29 and SW620 cells were separately treated with 80ug/ml 5-FU and 25ug/ml oxaliplatin for 8 hours. And apoptosis of cancer cells were analyzed and detected through AnnexinV-FITC/PI double staining flow cytometry technique. And proportion of CD133+ cell before and after chemotherapy were analyzed by flow cytometry being marked by CD133 antibody. Results: The following 10 cancer cell lines among the 42 cancer cell lines were detected as CD133 positive cells, such as SK-HEP-1, HepGII, Hep3B, L2-2-15 and PLC/PRF5 hepatoma carcinoma cell line, PANC-1, BXPC-1 pancreatic cancer cell line, SW620, HT29 colon cancer cell line, and CNE-2 nasopharyngeal carcinoma cell line. After separately treated with 80ug/ml 5-FU and 25ug/ml oxaliplatin, the proportion of apoptosis cells of HT29 and SW620 cells had been increased obviously, The proportion of the group treated with oxaliplatin increased higher than that of the group treated with 5-FU.The proportion of CD133+ cancer cells had been increased obviously. Conclusion: Among the 42 cancer cell lines, there are ten cancer cell lines were detected as CD133+ /TICs; It suggested that CD133 can be superficial filtration mark; And CD133 positive colon cancer cell have anti-chemotherapy activity obviously.
