Experimental Study On Treatment For Acute Gouty Arthritis By Regional Injection Of Colchicine

OBJECTIVE: To observe the therapeutic effects and side effects of intraarticular injection of colchicine for acute gouty arthritis.METHODS: 70 SD rats were divided into 7 teams in random。A:normal ;B:model control;C:oral colchicines(0.02mg/300g);D:intraarticular injection of colchicines(low dose,0.01mg/300g);E: intraarticular injection of colchicines(mid dose,0.02mg/300g );F: intraarticular injection of colchicines(high dose, (0.02mg/300g));G :intraarticular injection of hormone(0.4mg/300g) . Observed the general state ,arthrocele states of rats in every team, measured the perimeters and ankle joint arthrocele index. 72 hours after modeling, collected venous blood to perform leukocyte differential count and determine concentration of acidum uricum. Collected synovial fluid of right ankles, then observed sodium urate crystal under polarization microscope. Made mineral wax sections with right ankles removed from the rats for HE staining to observe inflammation of synovium under light microscope. RESULTS:①Acute gouty arthritis of rat's ankle was successfully induced by injecting 0.05ml sodium urate suspl (20mg/ml).②In group E, arthrocele states of rats were improved, total WBC and PMN count decreased, the total index of inflammatory cell infiltration, fiberoptic proliferation, synovail cell proliferation and edema congestion in synovial tissue diminished. 36 hours after intraarticular injection of colchicines , arthrocele of rat's ankle obviously lessened.Less rats in group E had symptoms of diarrhea and food-intake decrease,and there was significant difference compared with group C(P<0.05).CONCLUSION: Intraarticular injection of colchicine is a effective therapy of rats acute gouty arthritis. Mid dose had better therapeutic efficacy and less side effect.
