Objective: To study the correlation between the BMP-2 and MMP-9 and premature repture of membranes.Methods: Select 18 samples from each of the following groups– patients at term in labor ( the group at term in labor with intact membranes), patients at term premature rupture of membranes not in labor( the group at term PROM ), patients preterm labor ( the group preterm labor with intact membranes ), patients preterm premature rupture of membranes not in labor ( the group preterm PROM ) and patients at term not in labor (control group ); sample of chorioamniotic membranes from the rupture after the labor; manipulate immunobistochemistry and pathologic image multimedia operation system to conduct semiquantitative analysis of the area integrated optical density of the positive expression of BMP-2 and MMP-9.Results: (1) The AIOD of BMP-2 in chorioamniotic membranes of the group at term in labor, the group at term PROM, the group preterm labor and the group preterm PROM was 0.0137±0.0094,0.0269±0.0127,0.0273±0.0145,0.0456±0.0114 respectively. All the value was higher than that of control group which was 0.0060±0.0079 ( P=0.0068,P=0.0003,P=0.0004,P<0.0001). The BMP-2 of the group at term PROM was higher than that of the group at term in labor ( P=0.0065 ). The BMP-2 of the group preterm PROM was higher than that of the group preterm labor ( P=0.0069 ). The BMP-2 of the group preterm PROM was higher than that of the group at term PROM ( P=0.0037 ). (2) The AIOD value of MMP-9 in chorioamniotic membranes of the group at term in labor, the group at term PROM, the group preterm labor and the group preterm PROM was 0.0102±0.0126,0.0281±0.0161,0.0287±0.0145,0.0472±0.0118 respectively. All the value was higher than that of control group which was 0018±0.0033 ( P=0.0068,P=0.0005,P=0.0003,P=0.0002 ). The MMP-9 of the group at term PROM was higher than that of the group at term in labor ( P=0.0069 ). The MMP-9 of the group preterm PROM was higher than that of the group preterm labor ( P=0.0052 ). The MMP-9 of the group preterm PROM higher than that of the group at term PROM ( P=0.0065 ). (3) 17 patients out of the five group had histologic chorioamnionities, among which 13 patients were from the group with PROM ( 13/36, 36.1% ). The value of BMP-2 and MMP-9 in the membranes of patients with histologic chorioamnionities was 0.0201±0.0107 and 0.0200±0.0151 respectively, which was substantially higher that that of the patients without histologic chorioamnionities ( 0.0105±0.0092,0.0071±0.0084 )(P<0.05). (4) There was correlation between BMP-2 and MMP-9 expression in the membranes of all the five groups. ( rs =0.76159,P<0.0001).Conclusion:⑴The expression of MMP-9 and BMP-2 was increased in premature repture of membranes, which is closely relevant to the repture of membranes;⑵There was a correlation between BMP-2 and MMP-9 expression in chorioamniotic membranes.
