Study On Absorption Mechanism Of Rutacarpine

Modern Traditional Chinese Medicine (TCM) preparations are embodiments of the modernization of TCM. However, in order to modernize TCM preparations, it is more important to modernize the theoretical basis of dosage forms selected and the method for screening prescriptions, optimizing technics and evaluating quality than only to modernize pharmaceuical technique. Biopharmaceutics and Pharmacokinetics are subjects focusing on studying the regularities of drug's absorption, distribution, metabolism and elimination in vivo, and further elucidating the relationships between preparational factors, biological factors and drug effects. Thus, the purpose of this study is to provide novel ideas and approaches for selecting dosage forms, screening prescriptions, and optimizing technics by utilizing the latest measures and models of biopharmaceutics and pharmacokinetics.Rutacarpine, a complex TCM preparation, is widely used to treat coronary disease and gastrointestinal tract disease. In the present study, rutacarpine (RUT) was used as model drug. mutiple-model including rat in situ perfusion preparation and kinetic research in vivo was employed to investigate systemically absorptive mechanism and the best absorptive segment of the main componentss of RUT. There exist rather qualitative than good quantitative correlation between rat in situ perfusion model and in vivo kinetics investigation. In summary, more reliable conclusion may be achieved by utilizing two or more methods to examine the same drug and synthetically comparing and evaluating the results obtained from different methods.On the basis of preliminary conclusion of the absorptive mechanism about RUT, it was chosen as drug model for investigation the idea and method for the research of sustained-release praparation by the instruction of biopharmaceutics and pharmacok -inetics theory.Finally, the HPLC method, which determined the concentration of RUT in the Beagle dog plasma and could satisfy the need of pharmacokinetics study in Beagle dog, was established and method was initially certificated. Pharmacokinetics experiments of oral preparation RUT.In our study, absorption mechanism and influence factor of the extract of TCM was investigated using multi-model and multi-aspect method and biopharmaceutical and pharmacokinetical theory was also improved. The study also provided a new idea of RUT sustained-release dosage development, discussed a common pathway of the development and manufacture of the TCM sustained-release dosage, and explored the design, screening and establishment of quantity standard of extract of TCM sustained-release preparation formula by the instruction of biopharmaceutics and pharmacokinetics theory. The results and methods of this paper riches the contents of TCM sustained-release dosage, establish a good base of extract of TCM sustained-release dosage development, and greatly promote the modernization of TCM dosage.
