Alcohol is the major component of alcoholic beverage and is abused generally in the world. Over drinking may induce violence and accident. Blood alcohol concentration (BAC), which is primary gist dealing with affairs, need to be detected under many circumstances. After drinking, BAC will change from time to time. It correlates closely with alcohol pharmacokinetics, and alcohol pharmacokinetics correlates closely with alcohol dehydrogenase(ADH).We determine ADH activity in biological samples by HS-GC method, and investigate optimal conditions of ADH reaction, concentration and dosage of reaction stop buffer, related substances of alcohol. The optimal pH is 9.0.The optimal concentration of alcohol is 20.0mmol/L and NAD 10.0mmol/L. The optimal stop buffer is sodium dodecylsulphate (SDS).The pharmacokinetics of alcohol investigated in different group rats after oral administration of alcohol by method of HS-GC aboved. The result shows that comparing with blank group, pharmacokinetic parameter (K, Ka, Ke, Cl, Tlag) of experimental group rats are higher and other parameters (AUC, Cmax, Tmax) lower, parameters (t1/2Ka,compartment) not obviously change with drinking time.The ADH activity in serum, liver, stomach mucosa and intestine of experimental rats is determined and studied by statistics. The result shows as fellows, (1) ADH activity in stomach mucosa is descend, and it in liver rise, and it in intestine not obviously change. The ADH activity in serum rise also, but not obviously change in early days. (2) Both total-tissues and between-tissues comparison of different tissues in every groups is obviously different. The ADH activity in serum, liver, stomach mucosa of different groups is obviously different in total comparison, but it in intestine is not obviously different.The ADH activity is determined in serum of volunteers, drinking in short time comparing with drinking in longer time. And we arrange their data of alcohol pharmacokinetics that was already determined by our teams. The result shows that the change rules of ADH activity and pharmacokinetic data is similar with experimental group rats. These pharmacokinetic data with similar diversity are found. We study between it with ADH in serum of rat by correlating statistics. The result shows that there are closely correlated relationships between ADH in serum with K, Cmax, AUC, but not correlated with Ka. So under certain circumstance, activity of ADH in rat serum can obviously affect parameter of alcohol pharmacokinetic.The ADH activity is determined by HS-GC and terminal method. Both pharmacokinetic data of alcohol and ADH activity in model rats, and both their regularity are studied. The relationship that is between ADH activity in rat serum with pharmacokinetic data of alcohol is investigated primarily. These results can provide valuable reference for chronergy of alcohol in forensic practice.
