objective: In order to investigate the effect of tumor necrosis factor-αand pioglitazone on GLUT4 expression in 3T3-L1 adipocytes .Method: The differentiated 3T3-L1 adipocyte cells were divided randomly into three groups : the control group,incubated with TNFαgroup and incubated with pioglitazone group .The TNFαgroup was incubated with 20ng/ml TNFαand the pioglitazone group was incubated in 10-4mol/L pioglitazone,and after incubated with TNFαand pioglitazone respectively for 1d,3d,6d and then the expresstion of GLUT4 was determined .RT-PCR and Western-blot were used to detect the expression of GLUT4 gene and the experiment groups were compared with the control group.Results:RT-PCR and Western-blot analysis showed that TNFαtime-dependently reduce GLUT4 mRNA and protein in 3T3-L1 adipocytes and greatly decreased as compared with the control group ,and has statistical difference(P<0.01), while Pioglitazone time-dependently increased GLUT4 mRNA and protein as compared with the control group,and has statistical difference(P<0.05).Conclusion: TNFαand Pioglitazone have different effects on GLUT4 expression in 3T3-L1 adipocytes and have significantly statistical differences compared with the control group. Pioglitazone appeared to improve insulin sensitivity by increasing the GLUT4 expression and partly inhibit the effect of TNFαon insulin resistance in 3T3-L1 adipocytes .
