Aim To study the cardioprotective effect of recombinant human erythropoietin on lately reperfused rabbit ischemic myocardium. Methods 24 adult rabbits, weight range from 2.0 kg to 2.5 kg were randomly divided into 3 groups: Sham; Late Reperfusion (LR); Late Reperfusion with pretreatment of recombinant human Erythropoietin (E+LR). Each group has 8 rabbits. E+LR group was pretreated by intravenous administration of rhEPO (1000unit/kg) 12 hours before the operation. In reperfusion groups (LR,E+LR),coronary artery(Left Ventricular Branch, LVB)was ligated for 3h and then released for 3 h.Sham group were operated similarly without ligtation. All animals were sacrificed 6 h after the treatment. Blood was drawn from right ventricle for measuring the serum content of CK,LDH. Border regions of infarcted myocardium were incised and used for analyzing the tissue concentration of SOD,MDA,GR and the expression of FADD,Caspase-3 and the Apoptosis Index (AI). Results: Compared with the Sham group, the CK, LDH, MDA, FADD, Caspase-3 levels and AI were higher in E+LR group(all p<0.05) and SOD, GR were lower in E+LR group (all p<0.05).Compared with LR group , much lower CK, LDH, MDA, AI and much higher SOD ,GR were showed in E+LR group (all p<0.01). Conclusion: Pretreatment with recombinant human erythropoietin can protect the lately reperfused ischemic myocardium, which involves the mechanism of anti-oxidation and anti-apoptosis due to the block of activing pathway of Caspase-3.
