Objective: To build a model of rats suffered form hyperlipemia(HL) and severe acute pancreatitis(SAP).To investigate the change of gut mucosa permeability,gut epithelial cell apoptosis and proliferation of SAP and HL-SAP rats,we can understand something about the mechanism of gut barrier dysfunction on the rats of SAP.And we are to discuss the mechanism of guts barrier dysfunction in SAP and the cause of HL which aggravate SAP serious in hyperlipemia through detecting gut barrier dysfunction in HL-SAP rats.Method: Rats received intragastric administration of high fat diet 2 weeks which induced experimental hyperlipemia, and retrograde injection of 3.5% sodium taurocholate into the pancreatic duct which induced severe acute pancreatitis.50 male SD rats were randomly divided into four groups:control group,HL group,AP group,HAP group.After six hours when the models were completed, the histopathologic damage degree of pancreas were observed and the value of amylase(AMS),triglyceride(TG),cholesterol(CH) in serum were measured. Concentration of D-lactic acid and endotoxin shows the change of gut permeability.Terminal -deoxynucleotidyl transferase mediated nick end labeling( TUNEL)and Immunohistochemists were used to detect the cell apoptosis and expression of PCNA in the tissue of distal ileum.Results: The level of AP team is higher than than the controlled(p﹤0.01), pancreas is found to get bleeding and necrosis histopathologic change. HL`TG,CH in serum have get a higher leve(lp﹤0.01),the pancreas have no change. AP team which has higher D-lactic acid and endotoxin shows more apoptosis cell and lower expression of PCNA.HAP team get higher diet,more apoptosis cell and lower PCNA.Conclusion: A model of hyperlipemia rat with severe acute pancreatitis might be developed by intragastric administration of high fat diet 2 weeks and retrograde injection of 3.5% sodium taurocholate into the pancreatic duct. In the early stage of SAP, permeability become stronger,cell apoptosis and decreased proliferation may play important role here. Hyperlipemia can make SAP more serious through gut barrier dysfunction way.
