Objective:The research aims to plug into the role of infusion of Haploidentical cells radiated with 60Co in the cure of solid tumors to ease the GVHD and enhance the GVT effect and the changing of T cell excrete cytokine meaning on forecasting induce GVHD and GVT existing in the process through observing the clinical features of graft-versus-host disease (GVHD) and graft-versus-tumor effect (GVT effect) in the cure of hepatoma cells (H22) of F1 mices by infusion of Haploidentical cells respectively with and without radiation of 60Co among groups and check the changing belong to Th1 and Th2 cytokine in blood serum.Methods:Taking female CB6F1 mices (F1 mices in short) which have been hypodermically inoculated with hepatoma cells (H22) as recipients, after intraperitoneal chemotherapy we drew out the flesh marrow and spleen cells of the C57BL/6 mices as the donors which were divided into two groups with or without the radiation of 60Co. We divided the mices of F1 into 3 groups at random. We gave only received chemotherapy but no transplanting to group A. After giving chemotherapy to group B, we also infused the donor's cells which haven't been radiated by 60Co into their bodies. And group C received the cells which have been radiated by 60Co after given chemotherapy. We surveyed the performance of graft-versus-host disease (GVHD) showed by those three groups. We dyed the skin,intestines,liver and spleen of these mices from different groups on the seventeenth day after they received transplanting , sliced them and then surveyed the reaction caused by the changing of GVHD. Afterwards we inspected the weight of the tumor and the anti-tumor rate of different groups. On the second,fourth,seventh,eleventh and thirteenth day after transplanting, we drew out the retrobulbar venous blood from the rats of different groups, using the method of CBA to suspect the cell changing situation of IL-2,IL-4,IL-10,IFN-γblood serum.Results:1. The GVHD clinical performance of group C is apparently weaker than that of group B.2. Group C shows weaker changing of cell Pathology in the skin,intestines,liver and spleen than those of group B.3. The average weight of tumor of group A is (1.34±0.45)g, group B is (0.6±0.38)g, and group C is (0.28±0.15)g. Taking group A as a controlling group, we discovered the anti-tumor rate of group B is 55.22%, whereas group C is 79.22%。4. The cells of IL-2and IFN-Υwhich belong to Th1 in blood serum of group C is apparently decreased much more than those of group B, whereas the cells of IL-4 and IL-10 which belong to Th2 are increased.Conclusion:1.Haploidentical cells radiated with 7.5Gy 60Co as the donor's cells cause the GVT effect of H22-transplated mices with solid tumor and decrease the performance of GVHD. 2. Haploidentical cells radiated with 7.5Gy 60Co as the donor's cells can increase the cells of IL-2 and IFN-Υwhich belong to Th1 in blood serum of H22-transplated mices with solid tumor, whereas the cells of IL-4 and IL-10 which belong to Th2 are decreased.3. Days posttransplantation from four to seven , rise of IFN-γconcentration in blood serum of mices can denote enhancing effect of aGVHD .Days posttransplantation from four to seven, Rise of IL-4 concentration in blood serum of mices can denote reducing effect of aGVHD ,and signify restraining the effect of GVT。...
