Expression And Significance Of Activin A And Inhibin A In Preeclampsia And Eclampsia | | Posted on:2008-02-20 | Degree:Master | Type:Thesis | | Country:China | Candidate:L Jiang | Full Text:PDF | | GTID:2144360218956266 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | ObjectiveTo investigate the location and demi-quantitation of activin A and inhibin A in human placenta of preeclampsia and eclampsia,detect the level of them in amniotic fluid,compare the serum level of them before and after delivery,and discuss their roles in the pathogenesis of preeclampsia and eclampsia.MethodsImmunohistochemistry SABC and reverse transcription polymerase chain reaction(RT-PCR)were used to demonstrate the expression of activin A and inhibin A in women with preeclampsia and eclampsia(case group)and normal third-trimester pregnant women(normal group).The level of them in amniotic fluid, the serum level of them before and after delivery were detected by enzyme linked immunosorbent assay(ELISA).Results1.We found that activin A and inhibin A expressed in placenta obtained from case group increased compared with that in normal group,and had positive correlation with the disease.Immunohistochemistry demonstrated that they predominantly localized in trephocyte and endothelial cell of villous capillaries,The staining was stronger with aggravation of the disease.Both activin A mRNA and inhibin A mRNA had positive correlation with mean arterial pressure,alanine aminotransferase,aspartate aminotransferase,blood urea nitrogen,creatinine,uric acid,and had negative correlation with neonate body weight.Activin A mRNA had positive correlation with inhibin A mRNA in placenta.2.We found that serum activin A and inhibin A increased in case group compared with that in normal group before delivery and had positive correlation with the disease. Both activin A and inhibin A before delivery had positive correlation with MAP,ALT,AST,BUN,Cr,UA,and had negative correlation with neonate body weight.Activin A had positive correlation with inhibin A in serum before delivery.Serum activin A and inhibin A decreased after delivery.3.We found that activin A and inhibin A in amniotic fluid increased in case group compared with that in normal group and had positive correlation with the disease. Both activin A and inhibin A in amniotic fluid had positive correlation with MAP,ALT,AST,BUN,Cr,UA,and had negative correlation with neonate body weight. Activin A had positive correlation with inhibin A in amniotic fluid.4.We found that activin A mRNA and inhibin A mRNA in placenta had positive correlation with activin A and inhibin A in serum and amniotic fluid.5.We found that serum activin A and inhibin A before and after delivery in case group had positive correlation with blood pressure after delivery.The descending amplitude of serum activin A and inhibin A in case group had positive correlation with the descending amplitude of blood pressure.ConclusionThe expression of activin A and inhibin A in placenta,serum and amniotic fluid increased in women with preeclampsia and eclampsia.The placenta may be the main resource of them.Activin A and inhibin A may play an important role on preeclampsia and eclampsia by accommodating differentiation of the trephocyte.Serum activin A and inhibin A in case group had positive correlation with blood pressure after parturition.The descending amplitude of them had positive correlation with the descending amplitude of blood pressure.They could be used as the index to evaluate the prognosis of the disease. | | Keywords/Search Tags: | Activin A, Inhibin A, Preeclampsia, Eclampsia, Trephocyte | PDF Full Text Request | Related items |
