OBJETIVE To study the role of HIPK3 and Caspase-8 in the development of carcinoma of bladder, as well as its clinical significance, and to investigate the relationship between HIPK3 and Caspase-8 protein expression.METHODS The bladder cancer cell strain EJ was cultured, and induced to be apoptosis by mitomycin(MMC). We measured the rate of apoptotic cells to know the time-effect. Then the expression of HIPK3 and Caspase-8 was measured in immunohistochemical method. The more, we used FCM to research the influence of the antibody of HIPK3 and Caspase-8 to the cell apoptosis.RESULTS With the existing of MMC, cells' proliferation was inhibited. The apoptotic ratio was higher and higher when time passed. The expression of HIPK3 and Caspase-8 was masculine in the apoptic EJ cells, and the ratio was higher as well as time increasing. The ratio of Caspase-8 was lower than the HIPK3 in every corresponding time group. And there was no statistical difference in the ratio with the different dose of MMC, either. In SPSS, the masculine ratio of the two were positive correlation. The antibody of HIPK3 and Caspase-8 could oppose the effect of MMC, and decrease the apoptosis of EJ cells.CONCLUSION The up regulation of Caspase-8 expression in apopototic bladder neoplasm cells in connection with HIPK3 protein increasing reveals the close relationship between HIPK3 and Caspase-8, and the occurrence and progression of the cancer. |
