Objective:To analyze the mutation frequency of CCR5△32 and CCR5-894C deletion allele and the characters of polymorphism.Evaluation of the possibility of infection of those multi-nations residents,may provide foundations for anti-AIDS in theory.Materials and methods:Our analysis is based on 197 blood-samples of Mulao people from Luocheng Disease Preventing Office of Guangxi,100 blood-samples of Zhuang people and 100 blood-samples of Han people from Guangxi Medical University. The genome DNA was amplified by PCR method,we took 5μl PCR products from CCR5△32 to process 2%agarose gel electrophoresis.CCR5-894C deletion had been digested by HindⅢfor 2 hours,then 5μl products of which was taken to process 2%agarose gel electrophoresis.The results were observed and photographed under ultraviolet.The genotype of the products of PCR genome was judged according to the relationship between size and the amount of the products of PCR and the genotype.Then,one random portion of PCR products from Mulao,Zhuang and Han was purified with DNA Purification Kit. The purified PCR products then were ligated with pGEM-T Vector to construct recombinant plasmids,and translated into the cells of EPI-100.We selected those positive colonies with blue/white color screening on indicator plates and colonial PCR test,then defined the PCR products by sequence analysis. Results:1.No mutant of allele in CCR5△32 was found in those 397 units tested. All belong to the type of wild.2.No mutant of allele in CCR5-894C was found in those 397 units tested. All belong to the type of wild.Conclusions:After series of inspections in the frequency of mutant of HIV-1 co-receptor CCR5△32 gene and CCR5-894C deletion allele in Mulao,Zhuang and Han,no mutant was found.We can make a conclusion that the people of Mulao,Zhuang, and Han in Guangxi Autonomous Region face great genetic possibility in infection of HIV-1.During the past few years,the number of the infected in Guangxi has been soaring.Therefore,to pour extra supervision on HIV-1,and to foresee its trend,are quite necessary for us to prepare effective measures relatively.
