Objective: Synthesis appreciation quality of Saposhnicovia divaricata from different habitats, and to examine the most suitable harvest period of Saposhnicovia divaricata. To study the contents of chromones of Saposhnicovia divaricata of different parts and thickness and to dynamic analyze the active component of Saposhnicovia divaricata after Carex dispalatha.Method: The contents of four Chromone (prim-of-glucosy 1-cimifugincimifugin, 5-o-methylvisam, minal, sel-o-glu-cosy -hamaudol) were mensurated by RP-HPLC from different harvest periods, different parts (liber, hadromestome, cili-reeds, basal part of stem and root) of cultivated Saposhnicovia divaricata and different diameters in 11 different kinds of trueborn Saposhnicovia divaricata and 5 kinds of shoddy Saposhnicovia divaricata. The contents of moisture, ash, alcohol extractive and pesticide residue of heavy metal and organochlorine were Determined according to《chinese pharmacopoeia》set in 2005. The content of polycose which extracted by orthodox method (water decocting method) was mensurated by phenol concentrated sulfuric acid.Results.:(1)The mensuration results of Saposhnicovia divaricata from different habitats: The sample contents of moisture, ash, alcohol extractive measured up to the standard of《chinese pharmacopoeia》set in 2005. The total count of prim-of-glucosy 1-cimifugincimifugin and 5-o-methylvisam of ortho- product Saposhnicovia divaricata was super up to《chinese pharmacopoeia》set in 2005. Four chromone contents of different habitats Saposhnicovia divaricata had greatly difference (P<0.05). The highest was 0.86%(Antu, jiLin), the lowest was 0.35%(Nongxi, Gansu). The total count of four chromone of imitative Saposhnicovia divaricata was below 0.03%, and it was far lower than the limit value of《chinese pharmacopoeia》set.The contents of polycose of Saposhnicovia divaricata varied from 2.38% to 15.26%. There was greatly difference (P<0.05). Among them the contents of polysaccharose from AnTu, ChangLin of JiLin and XinBin of LiaoNing was 10% above. The content of AnGuo of Hebei was the lowest. The contents of H2O-solubility polycose of cultural orth- Saposhnicovia divaricata and hydro- Saposhnicovia divaricata(GanSu) were higher.The mensuration results of pesticide residue of heavy metal and organochiorine indicated the chromium content of eury- calices buchtorm libanotis root was 3.3-3.8 times to the contents of others, which were up to《Export and import Green vocation standardization of medicinal plant and praeparatum》.The contents of Fe,Mn,Sr and so on of grow wild Saposhnicovia divaricata were different to cultivante Saposhnicovia divaricata. Which could be used to identify grow wild and cultivante samples.Pharmaco- experiment of methanolic extract of Saposhnicovia divaricata manifested it's relieve fever effect was weak, but it's relieve pain and anti-inflammatory effect were significant. According to the correlative analysis of the contents of active component and methanol extract and pharmaco-active of methanolic extract, there were some correlation of the contents of chromones and methanol extract, but there were on correlation of the contents of chromones and the main pharmaco-active.(2)The determination results of different harvest periods: The total contents of four chromone of one year, two years, three years were 0.41%, 0.60%, 0.86% respectively. The best harvest year should be three years. About harvest month, the total content was highest on may, and second on October. Practical harvest period was often in autumn, so we should harvest them on October.(3)The determination results of different parts: The content of four chromone were 1.0989% in phloem, 0.2512% in hadromestome, 0.3678% in branch root, 0.3588% in small main root(3-7mm), 0.2387% in middle main root(7-10mm), 0.1795% in pachy main root(10-18mm), 0.009% in cili-reeds, 0.0932% in basal part of stem, 0.3048% in root. There were greatly difference (P<0.05) in them. It was thus clear that main active component of Saposhnicovia divaricata was centered in root.(4) Quality analysis results of Carex dispalatha: The content of four chromone was highest before Carex dispalatha in spring, and descended sharply after Carex dispalatha. The content of before Carex dispalatha was nearly 10 times to that after Carex dispalatha. For this reason, the Saposhnicovia divaricat of after Carex dispalatha was inadvisable to use as drug.Conclusion: The contents of four chromone and the index of routine test could be used to synthetic appraise Saposhnicovia divaricata. The quality of our country's cultural ortho- product Saposhnicovia divaricata was fine, and could be used as drug. The best quality of collected samples was Antu's and Xinbin's. The imitative Saposhnicovia divaricata couldn't used as drug to take the place of ortho- product Saposhnicovia divaricata. The main active component of cultivate Saposhnicovia divarieata had greatly difference in each part, and was centered in liber and root. To analyze thickness of root, the active component was main in rootlets and branch stems. We should think highly of them in practical application. But the concents of active component in cili-reeds and basal part of stem were extremely low. We should removal them in practical application. The contents of pesticide residue of heavy metal and organochlorine from different habitats had some difference, but they were all up to government standard.
