Lutein, a natural carotenoid, is widely distributed in fruits and vegetables, and is particularlyconcentrated in the Tagetes erecta flowers. Marigold flowers are ideal materials to extract lutein,because the lutein contents of marigold flowers are very high, and the contents of other categorycarrot pigment is extremely low. Marigold flowers are also the most important source of lutein forapplication in the food industry. The contents of free lutein in marigold is very low, lutein ester isits existing pattern, and it is not absorbed by bodies.However, the purity of lutein is requested tobe very high in multifunctional food additives. It is meaningful to expoilt functional lutein foodsin step with each other. As early as in the above century 80's intermediate stages, the westernmedicine researcher discovered that natural lutein is one kind of performance outstandingoxidation inhibitor. Lutein increases one quota to food in, may prevent a series of diseases fromthe human body caused by senile organ.The massive epidemiology evidence indicates, lutein hasthe protective function to the vision, and has effect on cataract, arteriosclerosis, enhancementimmunity and so on,specially play a emphatical role on preventing cancer from happening anddelaying cancer development, it is one hot spot on ingredient research of international functionfood in the present.There are little research on the lutein, and always in organic solvent extraction which wascomplicated. In order to solve the questions,we adopted ASE-100 Accelerated Solvent Extractor toextract the lutein, to optimize experiment parameters,and to establish technology method.Thetheoretical foundations and the practical value of the lutein extraction were provided for thedevelopment of the new food additives.We investigated the technogical conditions for extracting lutein from marigold withorthogonal experiment design proposal of five factors five levels,mensured the lutein content aftersaponifying by HPLC, and determined its optimum experiment parameters:extraction temperature80℃, dosage liquor ratio 1:30, Static time 10min,Static cycle three times, Flush volume 70%.The saponifying conditions of fatty lutein in marigold flowers in this experiment: saponifying temperature 70℃, saponifying times and saponifying alkaline density 40%KOH.We used marigold flowers powder as raw materials, extracting with 95% ethanol inultraphonic wave, and abstracting with organic solvent. After glucosans(Sephadex LH-20)laminar analysis, silica gel column chromatography eluting gradiently by acetidin, methanol andwater and purifying by silica gel column, we got five compounds totally which were extracted byTLC, MS and UV spectra and four of them were identified: quercetagein(Ⅰ),syringicacid (Ⅱ),quercetin (Ⅲ) andβ-amyrenol (Ⅳ).Compound(Ⅴ) hasn't identified, it's needed tostudy further.This text's new ideas: to extract lulein from marigold flowers first by ASE-100 AcceleratedSolvent Extractor.And to have determined its optimum experiment parameters with orthogonalexperiment by ASE-100 Accelerated Solvent Extractor.
