| Nowadays, the works on crude multiherb remedy are the hot spot of modernization of TCM. Crude multiherb remedy contain many complex chemical constituents, which is difficult and critical to study. Chromatographic multiple techniques play an important role in the analysis of multiplicity compounds. Crude multiherb remedy extraction is extremely complicate commixture, invariably containing many kinds of compounds, as well as inorganic substance, organics, small molecular, biomacromolecule, position isomer, enantiomer, polarity, nonpolarity, constant and trace materies ect; In the process of modernization of TCM, the study on chemical materials of crude multiherb remedy had proved to be an important and difficult work.According to the recent research on crude multiherb remedy and especially the defections on quality control and evaluation, the multi-dimensional fingerprinting of crude multiherb remedy and its evaluation methods were studied, with Xiaochaihu Tang as the subject. The determination methods of flavonoids compounds and saponins were acquired with two chromatograms to show two types of compounds with the different detected feature. UV detection got the determination results of flavonoids compounds and TOF/MS detection got the results of saponins. The method was firstly established to assay the six compounds and the pretreatment are easy and effective.Japanese scholars had reported that Xiaochaihu Tang and its analogical remedies (Xiaochaihu-Chaipu Tang, Xiaochaihu-Jiegengshigao Tang) could cause liver dysfunction, and it was supposed that hepatotoxicity should be relevant to Scutellaria baicalensis Georgi. Multiple linear regression were used to evaluate the TOF/MS detection fingerprinting and cell survival rate. The analysis results of linear regression equation indicated that the material of m/z 549.1591, molecular formula C26H28O13 might be hepatotoxicity element.The new-poietic and obsolescent compounds in Xiaochaihu con-decoction were studied. HPLC-TOF/MS fingerprinting and GeneSpring MS software could show the difference between crude multiherb remedy con-decoction and mon- decoction. By using the common chromatographic method, the TOF/MSD detection of finger- -printing of Xiaochaihu con-decoction and mon- decoction were obtained and Gene- -Spring MS software was applied to them. The new-poietic and obsolescent com- -pounds in Xiaochaihu con-decoction were found, which provided materies for tracing and interpreting crude multiherb remedy effection. Scientific connotation and chemical constituents of TCM compatibility of medicines in prescription were explored. |
