Array PDMS Microfluidics Toward Drug Metabolism Screening | | Posted on:2008-01-07 | Degree:Master | Type:Thesis | | Country:China | Candidate:B Chen | Full Text:PDF | | GTID:2144360218958957 | Subject:Drug Analysis | | Abstract/Summary: | PDF Full Text Request | | A microfludic system was developed for the analysis of P450-mediated metabolic reaction. The P450 enzymes were embeded in the channels of PDMS microfluidics. The metabolites by isozymes CYP3A4 and CYP2C9 were identified by LC-MS. This method has a few advantages over conventional analysis system, including analysis rapidity, decreased consuming reagents, the ability to recover and reuse the enzyme and ease of product isolation from the incubation mixture. We have applied HSCCC-HPLC-DAD for the first time to the preparative isolation and purification of hyperoside from Hypericum perforatum. A microfludic system was developed for the analysis ofβ-glucuronidase -mediated metabolic reaction. Theβ-glucuronidase enzymes were embeded in the channels of PDMS microfluidics. The hyperoside metabolited byβ-glucuronidase was identified by LC-MS. | | Keywords/Search Tags: | Hyperoside, Array microfluidics, PDMS, Drug metabolism screening, CYP450s, β-glucuronidase | PDF Full Text Request | Related items |
