| Objective: To study the expression of PTEN and CD44V6 protein in the rectal cancer, and the distance of mucosa adjacent to the rectal cancer. To discuss the relationship of the expression of PTEN and CD44V6 between the rectal cancer tissue and the mucosa adjacent to the cancer, major clinicopathological features was discuss too. This study was probe into PTEN and CD44v6 protein protect anus among the skill as cut reason predict value of factor far in carcinoma of the rectum..Methods: To examine the expression of PTEN and CD44V6 protein in the body of the rectal cancer , the mucosa adjacent to the cancer and normal colorectal mucosa. The relationship between the expression and the change of mucin. The immunohistochemical research help finish the study, and the samples came from the 28 patients who had got dissection for rectal cancer.The transitional mucosa adjacent to rectal cancer(1cm,3cm,5cm group) was the film dyed. Microscopic distal intramural expression was examined by PTEN/CD44V6-immunohistochemistry, HID-AB film dyeing,comparing with conventional hematoxylineosin staining in consecutive slice. Resulst:â‘ The positive expression of PTEN protein were 39.29%(11/28),39.29%(11/28),42.86%(12/28) ,89.29%(25/28) ,78.57%(22/28) in colorectal cancer,transitional mucosa adjacent to rectal cancer(1cm,3cm,5cm group) and normal colorectal mucosa respectively. Dealt with by statistics, the expressing rate in the former three are organized of PTEN protein, there is no significance difference (P> 0.05 ), in the 5cm group transitional mucosa adjacent to rectal cancer and normal colorectal mucosa group is no significance difference (P> 0.05 ) ,too. but The expression of PTEN protein in colorectal cancer and transitional mucosa adjacent to rectal cancer(1cm,3cm group) is significantly higher than that in normal colorectal mucosa,5cm group transitional mucosa adjacent to rectal cancer group (P<0.05). The positive expression of CD44V6 protein were 68.00%(19/28),64.29%(18/28) ,60.71%(17/28) ,24.29%(4/28) ,33.33%(7/28) in colorectal cancer,transitional mucosa adjacent to rectal cancer (1cm,3cm,5cm group)and normal colorectal mucosa respectively. Dealt with by statistics, CD44V6 protein is expressed in the former three are organized, there is no significance difference (P> 0.05 ), in the 5cm group transitional mucosa adjacent to rectal cancer and normal colorectal mucosa group is no significance difference (P> 0.05 ) ,too. but The expression of CD44V6 protein in colorectal cancer and transitional mucosa adjacent to rectal cancer(1cm,3cm group) is significantly higher than that in normal colorectal mucosa,5cm group transitional mucosa adjacent to rectal cancer group (P<0.05).â‘¡The expression levels of PTEN protein in colorectal cancer were significantly associated with the pathological classfications,TNM stage and Lymph node metastasis (P<0.05), The expression levels of CD44V6 protein in colorectal cancer were significantly associated with the invasion deep, TNM stage and lymph node metastasis (P<0.05).In colorectal cancer,the expression levels of PTEN and CD44V6 protein were moderately inversely (r=-0.592 P<0. 05).â‘¢From the transitional mucosa adjacent to rectal cancer(1cm,3cm,5cm group) was the film dyed, it were 5 samples who had had distal diffused, the ratio is 17.86%, the distance of the distal diffusion were all inner 1 cm.Other film dyed(HID-AB,PTEN,CD44V6), In all samples I had studied, the distance of mucosa adjacent to cancer were all inner 3cm,as well the transitional 3cm group mucosa adjacent to rectal cancer.â‘£In our study the positive expression of PTEN in transitional 3cm group mucosa adjacent to rectal cancer is 12 cases ,the positive rate is 42.86%,while the positive HID-AB film dyeing is 11, the rates is 39.29%.And the expression of CD44V6 is 17 cases, positive rate is 60.71% .There is no Correlation between the PTEN and HID-AB group or CD44V6 and HID-AB group respectively.So we must pay much attention to the transitional mucosa adjacent to the cancer.In the same time, the positive expression of PTEN and CD44V6 protein in transitional 3cm group mucosa adjacent to rectal cancer were moderately inversely (r=- 0.616, p <0.05).Conclusions:â‘ The positive expression of PTEN and CD44V6 protein in the rectal cancer and transitional 3cm group mucosa adjacent to rectal cancer is significantly higher than that in normal colorectal mucosa,transitional 5cm group mucosa adjacent to rectal cancer group (P<0.05).â‘¡PTEN and CD44V6 protein were associated with occurrence ,development and malignant degree in colorectal cancer. the expression levels of PTEN and CD44V6 protein were moderately inversely.â‘¢Conventional hematoxylineosin staining as 1cm distance of distal diffused of rectal cancer was not development, to the transitional mucosa adjacent to rectal cancer staining has PTEN, CD44V6 protein abnormal and the content of vitriol mucin decreased or disappeared.Suggest 3cm is safe domestically in the distal edge of rectal cancer dissection and its effect on anus-preserving operation. anus-preserving operation as a kind of radical cure operation, Patient with low degree or with late course of disease for splitting up , should consider the existence of the transitional mucosa adjacent to the cancer by the cancer , it is carried far excising the distance should not be confined to 3cm. |
