Objective: This paper was aimed to quantitatively reveal the chromatic andmorphologic characters of the exfoliate cells of lung cancer in sputum smears stainedby Pap test, to find highly specific parameters applied to differentiate and classify theexfoliate cells, and to discuss the value of the chromatic and morphologic parameterson classify and discriminate the lung cancer cells.Methods: 160 cases sputum samples were collected from the patients diagnosed withlung cancer and healthy peoples. The exfoliate cells in sputum smears were dividedinto normal and lung cancer groups. The normal groups include the cells of columnarepithelium (CE), squamous epithelium (SqE), histiocytes (HC). Each category has 20cases and 120 cells. The lung cancer cells were divided into squamous cell carcinoma(SqCa), adenocarcinoma (AD), small cell carcinoma (SCC) and large cell carcinoma(LCC) according to WHO classification. They consisted of 204 SqCa cells, 156 ADcells, 147 SCC cells, and 85 LCC cells. The sputum samples were smeared usingconventional method, fixed by 95ï¼…alcohol, stained by Pap test. The images of 7category cells were taken a photograph in objective lense 40X of light microscopeequipped digital camera and inputted into the computer. Technique of computerimage analysis was applied to test their relative parameters. The chromatic parametersincluded three primary colors of cell (Rc, Gc, Bc) and their coefficient (rc, gc, bc), three primary colors of nuclear (Rn, Gn, Gn) and their coefficient (rn, gn, bn). Themorphologic parameters of cells and nuclear included area (Ac, An), circumference(Cc, Cn), Long axis (dmaj,c, dmaj,n), Short axis (dmin,c, dmin,n), Axis Ratio (Rac, Ran),Area of cytoplasm (Acp), Karyoplasmic ratio (Rnp), Form factor PE (PEc, PEn), Formfactor AR (ARc, ARn), Regular form factor (RFFc, RFFn), and Form irregular index(FIIc, FIIn). The values of the parameters were tested with one-way ANOVA andcompared with Least—significant Difference. The following groups were submittedto do the stepwise discriminate analysis. They were the normal and lung cancergroups, four subtypes of lung cancer cells groups, and three categories of normal cellsgroups. And then stepwise discriminate functions were established, the effect ofdiscriminate was evaluated.Results:â… . The F value of these parameters in 7kind of intercellular comparisons respectively are 69.00, 32.23, 64.52, 22.98, 32.94,7.35, 70.57,54.94, 48.63, 53.24, 29.31, 21.57 after ANOVA, the P value is 0.000, thedifference has statistics significance.2. The value of Rc, Rn, Gc, Gn, Bc, rc, rn of Normal cell are higher than the lungcancer cell, that of bc, bn is lower than the lung cancer cell, the F value is respectively169.18, 49.59, 123.30, 28.55, 72.72, 62.75, 33.11, 67.13, 49.48, the P value is 0.000,the difference has statistics significance. The gn in between the normal cell and thecancer cell the difference, P value is 0.0723. In normal cell group, the values of Rc, Rn, Gc, Gn, Bc and Bn of SqE are highest,the values of Gc, Rn, Gn, Bc and Bn of CE are smallest, the value of Rc of HC issmallest, the F value is respectively 42.62, 11.90, 292.43, 52.36, 127.47, 27.23, the Pvalue is 0.000, the difference has statistics significance. Statistic test show significance amongdifferent ceil groups.Conclusions:1. Chromatic and morphologic features of exfoliate cell in sputum smearsstained by Pap test differ not only between the normal and the lung cancer groups, butamong the normal groups, and among the lung cancer groups. The difference reflecton the parameters Rc, Rn, Gc, Gn, Bc, Bn, rc, rn, gc, gn, bc, bn and Ac, dmaj, c, dmin,c, Cc,An, dmaj, n, dmin, n, Cn, Acp, Rac, Ran, Rnp, PEc, ARc, RFFc, FIIc, PEn, mRn, RFFn andFIIn. These parameters and their variance could be used to classify and discriminatethe exfoliate cells.2.
