With the aging society′s coming , osteoporosis is an important problem in the world . At present , many researches indicate that obstacle of calcium absorption in intestine in post-menopause is important in PMO . HRT can retrieve obstacle of calcium absorption in intestine , but it has adverse reaction . For example endome- trium bleeding , endometrium hyperplasia , Hormone replacement therapy (HR T)also can increase Gly , impact behavior of physiology and psychology result in brow ache and so on . So about 40-50 percent of American women reject to accept HRT . Acupuncture is effective and safe for treatment of osteoporosis without obvious side effect . The research of change of calcium absorption in intestine in post-menopause is very important for illuminating PMO′s etiopathog-enisis and i- nvestigating mechanism of osteoporosis of prevention and cure with acupuncture . ObjectiveWe determined OVX′s BMD and level of E2 in serum by means of DPX-Plus and ELISA to certificate therapeutic action of osteoporosis with acupuncture .We determined OVX′s expression of Calbindin 9k,1,25-dihydroxy vitamin D3 recep- tors mRNA in duodenal mucosa by means of RT-PCR in order to study regulation of calcium absorption in intestine with acupuncture .Methods1. Establishing of animal model50 heathy female rats were divided with means of randomly into five groups: normal,model,acupuncture,nilestriol,traditional medicine of Gusong Kang (GSKJN). Ten rats in each group. Anesthetized rats in groups of model,acupuncture,nilestriol and GSKJN were spayed.. One week after operation , rats in group of acupuncture treatment began to be acupunctured for 20 minutes every day. Rats in group of nilestriol treatment were administrated with nilestriol every day. The concentration is 50mg/L, every rat was administrated with 5ml nilestriol per week, other time every rat was administrated with 5ml normal saline; Rats in group of traditional medicine were administrated with capsule of GSKJN every day. The concentration is 120g/L, every rat was administrated with 5ml GSKJN per day;Rats in group of normal and model were administrated with normal saline at the same time, every rat was administrated with 5ml normal saline per day. Each group of rats eat normally, drink freely. After 8 weeks of intervention, all of the rats were to put to death.2. Detection of femoral bone′s BMDWe took out of rat′s left femoral bone , and detect rat′s femoral bone′s BMD by means of DPX-Plus .3. Detection of the level of E2 in blood serumWe got blood by the means of culling rat′s eyes for about 6 to 10 ml, and then segregated serum by means of centrifugalization. We determine OVX′s level of E2 in serum by the means of ELISA .4. Detection of OVX′s expression of Calbindin 9k,1,25-dihydroxy vitamin D3 r- eceptors mRNA in duodenal mucosaWe took out of rat′s duodenal mucosa, and the OVX′s level of Calbindin 9k,1,25-dihydroxy vitamin D3 receptors mRNA in duodenal mucosa were determined by means of RT-PCR .Results1. WeightCompared with groups of normal,acupuncture,nilestriol,traditional medic- ine of GSKJN, rat′s weight in model group increases significantly(P<0.01), there was not significant difference between the other four groups (P﹥0.05).2. Femoral bone′s BMDModel group compared with normal group,acupuncture group,nilestriol grou- p,traditional medicine of GSKJN group, rat′s femoral bone′s BMD decrease sign- ificantly( P<0.01) , the other four groups has not significant difference(P﹥0.05).3. The level of E2 in serumModel group compared with normal group,acupuncture group,nilestriol grou- p,traditional medicine of GSKJN group, rat′s level of E2 in serum decrease signi- ficantly(P<0.05); nilestriol group,traditional medicine of GSKJN group compare- d with normal group, rat′s level of E2 in serum decrease significantly(P<0.05); ni- lestriol group and traditional medicine of GSKJN group has not significant differ- ence(P﹥0.05); normal group and acupuncture group has not significant differen- ce(P﹥0.05).4. Expression of Calbindin 9k,1,25-dihydroxy vitamin D3 receptors mRNA in d- uodenal mucosaModel group compared with normal group,acupuncture group,nilestriol group, rat′s expression of Calbindin 9k mRNA is weaker, normal group,acupuncture group,nilestriol group has not significant difference; Model group compared w- ith normal group,acupuncture group,nilestriol group, rat′s expression of 1,25- dihydroxy vitamin D3 receptors mRNA is weaker, acupuncture group,nilestriol group compared with normal group, rat′s expression of 1,25-dihydroxy vitamin D3 receptors mRNA is weaker, acupuncture group,nilestriol group has not significa- nt difference. ConclusionAcupuncture could increase OVX rat′s BMD,the level of E2 in serum,the e- xpression of Calbindin 9k,1,25-dihydroxy vitamin D3 receptors mRNA in duo- denal mucosa, and decrease OVX rat′s weight. The result indicates that acupuncture could treat OVX rat′s osteoporosis by the means of up-regulation of expression of Calbindin 9k,1,25-dihydroxy vitamin D3 receptors mRNA in du- odenal mucosa to increase calcium absorption in intestine .
