AIM: Explore the mechanisms of Jiaomu Oil which is one of traditional Chinese medicine treating asthma of asthmatic model in guinea pig and provide theoretical basis for treatment of asthma with jiaomu Oil.METHODS:(1)Construct asthmatic model in guinea pig and divide 4 groups at random: normal control group, asthmatic model group, dexamethasone treatment group and Jiaomu Oil treatment group.(2)Measure airway hyperresponsiveness(AHR) of guinea pigs in every group. Then observe the treatment effects of Jiaomu Oil and dexamethasone on AHR of guinea pigs.(3)Determine the levels of IL-4 and IL-5 in serum of guinea pigs using enzyme-linked immunosorbent assay(ELISA).Then observe the treatment effects of Jiaomu Oil and dexamethasone on the levels of IL-4 and IL-5 in serum of guinea pigs.(4)The levels of IL-4 and IFN-γin BALF of guinea pigs were determined using ELISA. Then observe the treatment effects of Jiaomu Oil and dexamethasone on the levels of IL-4 and IFN-γin BALF of guinea pigs.(5)Observe the treatment effects of Jiaomu Oil and dexamethasone on the expression of NF-κB in the lung tissues of guinea pigs by immunohistochemical staining.RESULTS:(1)The PC20 of Jiaomu Oil treatment group[(0.160±0.180) g/L] was higher significantly than that of asthmatic model group[(0.013±0.014)g/L](P<0.05), so the AHR of the former was lower than that of the latter significantly.(2)The levels of IL-4 and IL-5 of Jiaomu Oil treatment group [(22.8±3.5)ng/L and (89.2±38.7)ng/L] were both lower significantly than those of asthmatic model group[(38.2±3.4)ng/L and (344.4±21.8)ng/L](P<0.05).(3)The level of IL-4 in BALF of Jiaomu Oil treatment group[(15.35±4.28)ng/L] was lower significantly than that of asthmatic model group[(41.36±6.71)ng/L](P<0.05).The level of IFN-γin BALF of Jiaomu Oil treatment group[(50.65±4.19)ng/L] was higher significantly than that of asthmatic model group[(21.15±2.75)ng/L](P<0.05).(4)The percent of NF-κB positive cells in Jiaomu Oil group [(29.89±2.01)%] was lower significantly than that of asthmatic model group[(52.71±7.80)%](P <0.05).CONCLUSIONS:(1)The mechanisms of Jiaomu Oil which could decrease the AHR effectively may be that it could prevent the secretion and releasing of IL-4 and IL-5 in serum of asthmatic guinea pigs.(2)Jiaomu Oil could decrease the level of IL-4 efficiently and increase the level of IFN-γin BALF of asthmatic guinea pigs in order to keep the balance of Th1/Th2 cells.(3)Jiaomu Oil could decrease the infiltration of inflammatory cells to lung tissues and the expression of NF-κB in the airway of asthmatic guinea pigs.In summary, Jiaomu Oil could treat asthma of asthmatic model in guinea pig.
