Hepatocyte growth factor (HGF) is a polypeptide GF. It is first acquired from the serum of rat suffering from part liver ablation. It can encourage many kinds of cells'accretion, deadexis, morphogenesia (including hepatic cell, epithelial cell, endothelial cell, hematopoietic cell and so on)and participate their generation, deadexis. HGF has strong induction in various tumors'infestation and aversion. At first, it was considered one of the mitogens,.later we founded that it has many effect on tumor's accretion. HGF'S BA is mediate by proto-oncogene c-met and it has strong induction. After HGF and c-met binding together,the recepter start autoallergic phosphorylation and activate it's tyrosine kinase activation, cascade activate a series of signal transduce path , and then accommodate cell's generation, differentiation, motion,vascularization.HGF/c-met expressed in ovario tumor cell are close correlate with MVD. .HGF/c-met expressed high cause it's sensitiveness to HGF,and made the cell excessus hyperplasied, differentiated and aggravated.C-met protein, the receptor of HGF, was distinguished in 1991.It is independence phosphorylation transmembrane receptor. It comes from the precursor protein (mr190000)'s desintegration. C-met is made up byαsubunit (50kd) andβsubunit (145kd), it is a heterodimers and has three different structural domain, extracellular region, transmembrane domain and intracellular region.αsubunit andβsubunit's extracellular region discriminate and comb HGF as the recognition site. In tracellular region has PTK activity. Prat founded that some cancer cell's met precursor (mr190000) are lack of schizolysis, it already has activity without connect ligand, and lose the accretion of HGF.The accretion and aversion of tumor is depend on the production of blood vessel .it will form duplicated and deadexis has intensify. HGF is a strong inductor of neogenesis blood vessel ,it can induce endothelial cells generation and extension, irritate the growth of new blood vessel. For all above, we decide to investigate the expression of HGF/c-met in ovarian cancer cell, try to analysis HGF/c-met signal conduct in ovarian cancer infestation and diversion.PURPOSE: 1. To identify the relationship between the expression of HGF/c-met extent and ovarian tumor malignancy level.2. To identify the realationship between HGF/c-met and MVD.3. To identify the effect of HGF/c-met on ovarian tumor cell infestation and diversion.4. Try to discuss the relationship between HGF/c-met and taxinol's mechanism of action.METHOD:1.Immunohistochemistry method was used to detect the expression of HGF/c-met in human ovarian tumor.2. RT-PCR,in sity hybridization was used to detect the expression of HGF/c-met in human ovarian tumor cell ( in vitro culture).3. Using Transwell to detect HGF/c-met influence on ovarian tumor cell infestation and diversion.4. Cell culture and MTT method was used to made growth curve (surely color taxinol intervention); electron microscope was used to observe the change of expression.RESULT:1. Ovarian tumor can be detected the expression of HGF/c-met mRNA, and they close correlate with tumor's effluence, infestation and infestation. 2. Human ovarian tumor cell (in vitro culture) can be detected the expression of HGF/c-met mRNA. A certain concentration of HGF can regulate the expression of c-met mRNA up.3. Transwell cultivate 24h, use fibronectin to made cells which were breakthrough the membrane, enumeration with microscope after staining. We found that HGF/c-met can enhance tumor cell's infestation.To observe with scanning electron microscope, HGF/c-met express heavily on both cell nucleus and cellular, in another group, we add invariably color taxinol and we found its expression has been hold-back, the excitat voltage which was need by display the figure h as rise, too.CONCLUSIOU:1.The expression of HGF/c-met existed in ovarian tumor and they are correlatively. They have close relationship with tumor's effluence, infestation and infestation.2.HGF/c-met can remarkably improve the infestation ability of ovarian tumor cell.3.To inhibit the expression of HGF/c-met may be a pathway of taxinol's lethal effect on tumor.
