| Objective To investigate IFN-γ,IL-4 production of peripheral blood mononucler cells in response to CpG ODN+ HBsAg in acute and chronic hepatitis B infection. Methods Random collect forty four patients chronically infected with HBV,18 patients in acute HBV recovery phase.The peripheral blood mononuclear cells(PBMCs)from the patients separated by density gradient centrifugation were stimulated with CpG ODN,HBsAg,CpG ODN+ HBsAg separately in vitro,compared with cells stimulated with 1640.The production of IFN-γ,IL-4 was measured by method of enzyme link immune spot.Results PBMC stimulated with CpG ODN+ HBsAg,HBsAg in acute and chronic hepatitis B,the secretion of IFN-γobviously higher than blank,there is statistics difference(P<0.021).PBMC stimulated with CpG ODN+ HBsAg,HBsAg in acute HBV recovery phase and chronic hepatitis B( ALT80-200 U/L),the secretion of IL-4 obviously higher than blank,there is statistics difference(P<0.028).PBMC stimulated with HBsAg,HBsAg+CpG in acute HBV recovery phase,the secretion of IFN-γobviously raise than chronic hepatitis B,there is statistics difference(P<0.005).PBMC stimulated with HBsAg,HBsAg+CpG in chronic hepatitis B group(ALT80-200 U/L),the secretion of IFN-γobviously raise than the others chronic hepatitis B (ALT<80U/L or ALT>200 U/L), there is statistics difference(P<0.005). Conclusions Stimulate with CpG ODN+ HBsAg,HBsAg,the production of IFN-γraise significantly in acute and chronic hepatitis B patients. PBMC stimulated with CpG ODN+ HBsAg,HBsAg in acute HBV recovery phase,the secretion of IFN-γobviously higher than the patients in chronically infected with HBV. PBMC stimulated with CpG ODN+ HBsAg,HBsAg in chronic hepatitis B (ALT80-200 U/L),the secretion of IFN-γobviously higher than the other groups. CpG ODN+ HBsAg can efficiently enhance acute and chronic HBV patients immune response,it prompt CpG ODN may be use in therapy HBV infection as immunomodula in future. |
