We obtained seven polyclonal antibodies (PcAb) against rat albumin by immunizing rabbits and sheep. The polyclonal antibody from No. 2 sheep had better ability. It was fit for the development of kits.We developed RIA and ELISA kits for rat albumin. The measurement range of the assays was 1~50μg/ml, sensitivities of the assays were 0.12μg/ml and 0.42μg/ml, recoveries of rat albumin were97.8~108. 4% and 85.0~106.0%. lntra- and inter-assay variation coefficients of radioimmunoassay were 3.2% and 6.9% respectively. Intra- and inter-assay variation coefficients of ELISA were 7.6% and 8.9% respectively. The correlation coefficients between measured and expected values were more than 0.990 after serial dilution of the rat urine samples with high concentrations of rat albumin. The value for normal rat urine samples (n=20) was 0~21.15μg/ml, Compared the RIA with the ELISA, the correlation was y=0. 92x+0. 08, r=0. 976, n=44. The kits for rat albumin might provide a convenience in exploitation of renal drugs and experimental injury of the kidney.
