| Objective To examine the expression and clinical significance of Smac gene in epithelial ovarian tumors. Methods The semi-quantitative RT-PCR method was used to detect the relatively quantity of Smac mRNA in 25 patients with malignant ovarian tumors,10 with borderline ovarian tumors,20 with benign ovarian tumors and 20 cases of normal ovarian tissues; Western blot was used to detect the expression of Smac protein. The same methods were used to observe the expression of Smac mRNA and protein in the ovarian carcinoma cell line A2780 and it's cisplatin resistant cell line A2780/DDP. Results Expression of Smac gene decreased significantly in malignant and borderline ovarian tumors when compared with those in normal ovarian tissues and with benign ovarian tumors. The expressions of Smac mRNA and protein in A2780 cell line were much more than those in A2780/DDP cell line. Conclusion Downregulated expression of Smac gene may play a role in the development of ovarian cancer,and also in the mechanism of drug resistance. |
