Objective: To investigate the changes and potential roles of Toll-like receptor 2 and 4mRNA expressions in lungs in acute hemorrhagic necrotizing pancreatitis(AHNP) rats.Methods: Seventy SD male rats were randomly divided into Sham-operated group(n=10), AHNP group(n=30) and Chloroquine-treateded group(n=30). AHNP was induced by infusion of 5%STC(0.1ml/100g) into bilio-pancreatic duct. Models of sham-operated were flipped ceca. Rats of AHNP groups and CQ-treated groups were sacrificed at 3,6,12 hours; sham-operated group were sacrificed at 6 hours. Samples of lungs and blood were taken for analysis.The lungs were dissected for lung histological scores and bronchoalveolar lavages were harvested for lung injury index. TLR2/4mRNA expression in the lungs were measured by RT-PCR.Results: TLR2/4 mRNA could be detected in lungs with low values in sham-operated group(0.016±0.664E-2, 1.003±7.192E-2), but they were markedly increased at 3 hours in AHNP group(0.787±2.376E-2, 1.512±5.674 E-2), peaking at 12 hours(1.113±0.108, 2.957±8.284E-2; P<0.05) . Lung injuries were aggravated, the levels of TNF-αmRNA expression in lungs were increased and the level of NO in lungs were decreased(P<0.05). Treatment with CQ could effectively inhibit TLR2/4mRNA expression(3h: 0.333±0.175, 0.536±0.2196; 6h: 0.488±0.215, 0.928±0.2958; 12h: 0.883±0.262, 1.840±0.6004; P<0.05) and relieve lung injuries. The levels of TNF-αmRNA expression in lungs were decreased and the levels of NO were markly inreased(P<0.05) .Conclusion: These data suggest that expression of TLR2/4mRNA is increased in lungs in AHNP. And up-regulation of TLR2/4mRNA expression in lungs may be involved in the genesis and the development of acute lung injury complicated in AHNP.
