Matrix Metalloproteinases-9 (MMP-9) is related with human autoimmune disease and plays a key role in the pathogenesis of these diseases ,such as rheumatoid arthritis, primary sj?gren's syndrome,multiple sclerosis.It has been reported that the high lever of MMP-9 in SLE patients'serum ,but the expression of MMP-9 on peripheral blood nuclear cells(PBMCs) of patients with SLE has not the report in our country.PBMCs include T lymphocyte,B lymphocyte and monocyte.They are all the great immunoregulation cells in human body. The study is to determine the expression of MMP-9mRNA and TIMP-1mRNA of PBMCs in SLE patients and healthy controls.Objective To determine the expression of Matrix Metalloproteinases-9 (MMP-9)mRNA and tissue inhibitor of metalloproteinase-1(TIMP-1)mRNA of peripheral blood nuclear cells(PBMCs) in patients with systemic lupus erythematosus(SLE) ,and analyze the relationship between the MMP-9 and TIMP-1 of PBMCs and SLE activity. Methods The levers of MMP-9mRNA and TIMP-1mRNA of PBMCs in patients with SLE and healthy controls were measured by reverse transcription-PCR (RT-PCR) .Results The expression of MMP-9 mRNA of PBMCs in active SLE patients (n=22 ) was significantly high compared to inactive patients (n=17) and healthy controls(n= 15), The expression of MMP-9 mRNA of PBMCs in inactive patients was significantly highter than that in healthy controls (P<0.01), the expression of MMP-9 in patients proteinuria was significantly high compared to those without proteinuria (P<0.01),the lever of MMP-9mRNA of PBMCs had positive correlation with SLEDAI scores ((P<0.01).There were no significant differences of PBMCs TIMP-1 expression between different groups (P>0.05), and the lever of TIMP-1 had no correlation with SLEDAI scores (P>0.05).Conclusion The expression of MMP-9 mRNA is high in PBMCs from patients with SLE and its level is potively related to disease activity ,which may reflect the important role in the pathogenesis of SLE.
