Extraction, Seperation And Protective Effect On Liver Of Triterpenoids In Artemisia Selengnesis Turcz.

The extraction, identification, separation and liver protection of triterpenoidsfrom Artemisia Selengnesis Turcz. were investigated. Then, Liver protection Functionof triterpenoids from Artemisia Selengnesis Turcz. was reaearched.Firstly, Triterpenoids in Artemisia Selengnesis Turcz. were determined byqualitative test. Then, oleanolic acid was identified by thin layer chromatographymethod, and it is contained 52.7% in leaves which were extracted by cyclohexane bymeans of the analysis of peak areas of HPLC.Secondly, Being optimized the process of colorimetric method which used todeterme total triterpene contents. Method validation showed that colorimetric methodwas feasible.Thirdly, Important factors and levers were determination by single factorexperiment,orthogonal experiment and response surface experiments. According toresult, influence factors sequence arrange that concentration of ethanol＞extractiontemperature＞liquid-solid ratio. Experimental model was typicalness analyzed bysoftware of Design-Expert 6.0.5 and the best process conditions of extraction as:concentration of ethanol was 87%, liquid-solid ratio was 9.8:1, extraction temperaturewas 74℃. 0.36% of dry weight of Artemisia Selengnesis Turcz. is contributed bytotal triterpenes and 41.5% contributes to ethanol extract.Fourthly, Optimized the process of thin layer chromatography,silica columnchromatography and HPLC.Finally, Protective action of triterpenoids on cheraical by tests of lowering theactivity of ALT in serum in vitro, free radical scavenging, anti-lipid peroxidation andbacteriostatic experiment. Functional component,technology,health function wereprovided experimental basis for development of health food.
