Natural products provide the new structural, curative effect and low toxic compound for drug research. This dissertation was consisted of two parts:The first content contained the separation, purification, structure conformation and the test of biological activity.1. The KB paper diffusion method was performed to screen the antibacterial activity of the rude component of the Myrica rubra kernel, and the pure compounds were bioactivity-guided separated. The compound separated was investigated according to antibacterial activity.2. The fatty acids and waxes of Myrica rubra kernel were analyzed through GC-MS, The results showed that the yield of kernel oil was 31.10%, and there were eight fatty acids and three long-chain alkanes in the extracted composition. The main composition were Palmitic acid 28.22%, Linoleic acid 14.4%, Oleic acid (Z) 25.74%, Erucic acid 15.68%, Heptadecane 5.2%, Octadecane 5.87%, Nonadecane 4.71%. The total content of unsaturated fatty acid was 55.91% with the Linoleic acid and Oleic acid of 40.19%, these supplied a reference to its comprehensive utilization.3. The Myrica rubra kernel were obtained from Tonghai Food Group Ltd. Company of Zhejiang, and were dried on the condition of light-free, powdered and extracted with 99% alcohol. The dissertation gave antibiotic activity screen according to their polarity and trail-separated these compounds. The compounds were isolated by repeated column chromatography (CC) on silica gel, RP-18, and Sephadex LH-20, as well as by preparative thin layer chromatography (PTLC) and recrystallization. The structures of these compounds were elucidated by means of spectroscopic methods including IR, UV, EI-MS, FAB-MS, HR-ESI-MS, 1D-NMR (1H-NMR, 13C-NMR, DEPT and NOEDS) and 2D-NMR (1H-1H COSY, HMBC, HMQC and NOESY). From Myrica rubra kernel, twenty-two compounds were isolated and sixteen of them were structurally elucidated, the compounds were flavones YM-6 e.g., triterpenes YM-16 e.g. and steroids YM-1 e.g.4. The compound obtained from Myrica rubra kernel was studied on its antibiotic activity. The compound YM-16 had antibiotic activity to Stenotrophomonas maltophilia and Escherichia coli. The similar antibiotic activitiys on Stenotrophomonas maltophilia were shown at the concentration of 15 mg/mL of the compound YM-16 and of 30 mg/mL of ancient Litton chlortetracycline, with the bacteriostasis circles diameter of 7.1 mm and 7.3 mm.5. EGCG abound in Myrica rubra kernel was first hydrolysated into EGC, and the conditions of reaction was optimized.The second content contained the separation, purification, constructer conformation and activity test of chemical composition in the secondary metabolite of marine fungus Fusarium sp..1. This dissertation investigated secondary metabolite of Fusarium sp., and primarily screened the untituberculosis and hepatotoxicity activity of the compounds obtained.2. Fusarium sp was obtained from Spartina alterniflora .This dissertation fermented and cultivated fungus ,and extracted the mycelium and fermentation broth with 99% alcohol. The two solvents were dried evaporated under reduced pressure and the residues were tested through HPLC, and discovered that the content in two residues were similar.The two residues were combined and 12 compounds were obtained from them through repeated column chromatography (CC) on silica gel, RP-18, and Sephadex LH-20, as well as by preparative thin layer chromatography (PTLC) and recrystallization. The structures of 8 compounds, such as alkaloid F4-3 e.g. and steroid F4-1, were elucidated by means of spectroscopic methods.3. The untituberculosis activity of these compounds was tested through two tube dilution method, and the toxicity to healthy hepatocell was investigated with MTT. F4-3 showed some activity aginst Mycorbacterium tuberculosis H37RV, the value of MIC100 was 5μg/mL, and little toxieity to healthy hepatocell.
