Photodynamic therapy(PDT)was a new therapeutics to cure cancer by photodynamic action,and had more obvious advantages compared with other traditional treatments. Lamp-house,photosensitiser,the oxygen concentration and the type of cancer cells were the main influencing factors in PDT.It was very important to choose appropriate lamp-house to excitated photosenzitisers and decided the effection of PDT.Photodynamic therapy was used to cure entity tumor initially,and it was used for ex vivo purging of leukemia cells to destroy residual cancer cells in bone marrow autotransplant treatment after 1980's.In this paper,basic mechanism of PDT was introduced simply at first,and then actuality of PDT for ex vivo purging of leukemia cells was described.The shortage of investigations presently was pointed out,the purpose and significance of this paper were presented.5-aminolevulinc acid based photodynamic therapy(ALA-PDT)experiments for ex vivo purging of leukemia cell K562 is the mostly research of this paper.In the progress of research,several basic experiment conditions were studied and the optimal experiment parameters were chose.Lamp-house,used to excitated photodynamic action in ALA-PDT, was researched mainly.Conclusions got from experiments as follows:1.On condition of 10~5cells/ml in culture medium,the optimal photosensitiser dose was 1mmol/L,and the optimal culture time was 4 hours in the ALA-PDT experiment to destroy K562 cells.2.The quantity relation between effect factors,in the process of K562 cells destroyed by ALA-PDT,was confirmed.It's a useful reference method for other photodynamic therapy experiments and clinic application.3.The inhibition rate of K562 cells caused by different Lamp-houses excitated ALA-PDT was analyzed.The Xe lamp was chose as the optimal lamp-house in ALA-PDT. In this experiment,the optimal parameters got from prior experiments were used.
