PrefaceFunctional dyspepsia(FD) is a common syndrome. Studies have shown the effect of Chinese herbs on gastrointestinal motility as well as treatment of FD clinically. In this researchwe analyzed the interaction of each component herb in Xiaoshiheweichongji, which was made on the basis of Baohewan, by the method of orthogonal design, to certify rationality of this herb, and its influence on gastrointestinal motility.ObjectiveTo investigate the effect of Xiaoshiheweichongji on the gastrointestinal motility of normal and dysmotility rats by the method of emptying nutritional semi-solid feeding; To study the interaction of each component herb in Xiaoshiheweichongji,and confirm its rationality.Methods1. The influence of Xiaoshiheweichongji on gastrointestinal motility of normal mice: Kun Ming mice were randomly allocated to control group, Mosapride, Xiaoshiheweichongji, Baohewan respectively by oral everyday during a period of 7 days, and were fasted an overnight before study.We used nutritional semi-solid as the gastrointestinal transporting marker to observe the rate of the residue in stomach and small intestinal propulsion rate after 30 minutes giving mice makers by oral. 2. The mice's models of gastrointestinal dysmotility: The mice were divided into controlled group and model group. The model group was injected into abdominal cavity with L-Arg(the dosage of the first day was 7.4g/Kg, of the second to fifth day was 3.7g/Kg), the controlled group was injected with saline of the same dosage. On the sixth day, we put the mice to death without feeding for 18 hours after being poured the nutritional semi-solid paste for 30 minutes and measure the rate of residue in stomach and the rate of the small intestinal propulsion.3 . The influence of the xiaoheweichongji on the mice's model of the gastrointestinal dysmolity induced by L-Arg : to investigate the effects of Xiaoshiheweichongji, Mosapride and distilled water poured into the mice model of gastrointestinal dysmotility induced by L-Arg.4. To screen the effect and interaction of each component herb on gastrointestinal motility of normal mice: We used the method of orthogonal design list L16(215) to determine the degree of role each component herb in the compound herb, and the mutual roles of them. There were 7 factors and each factor had 2 levels, level 1 standing for the concentration of herb being 0, level 2 being certain concentration of each herb. We used different decoctions based the level of main effect of the orthogonal design L16(215) to make 16 experiments.Results1. Effects of Xiaoshiheweichongji on gastrointestinal motility of normal mice: compared with the control group: the significant effect of Xiaoshiheweichongji promoting gastric emptying of normal mice poured with nutritional semi-solid was similar to Mosapride, but advantage to Baohewan, which also could play a significant role on gastric emptying and small intestinal propulsion.2. Effect of L—Arg on gastrointestinal motility: L—Arg coule inhibit gastric emptying and small intestinal propulsion significantly(P<0.01).3. Effect of Xiaoshiheweichongji on mice's model: Xiaoshiheweichongji could improve the gastric emptying and small intestinal propulsion of the mice's model of the gastrointestinal dysmolity induced by L—Arg significantly, and its effect was found to be close to Mosapride. However, the effect of both Xiaoshiheweichongji and Mosapride had no statistical significance on the recovery of gastrointestinal motility(P>0.05).4. Effect of each component herb on gastric emptying: Based on the analysis of orthogonal design, we conclude that the gastric emptying rate of seven component herbs increased: Fructrs Amomi, Fructus Crataegi, massa medicata fermentata, Fructus aurantii immarurus. The others had the opposite effects. As the analysis of variance showed, Fructrs Amomi, Fructus Crataegi and massa medicata fermentata could promote the small intestinal propulsion significantly; Semen Raphani and Glycyrrhiza had little effect, and Poria, however, could inhibit gastric emptying. As a result, the combination of Fructrs Amomi and massa medicata fermentata, Fructus Crataegi and Semen Raphani, Fructus Crataegi and Fructus aurantii immarurus could promote gastric emptying by significant synergism.5. Effect of each component herb on small intestinal propulsion: Based on the analysis of orthogonal design, we concluded that the rate of small intestinal propulsion of seven component herbs increased: Fructrs Amomi, Fructus Crataegi, Semen Raphani, Fructus aurantii immarurus; Glycyrrhiza and Poria had the opposite role. As the analysis of variance showed, Fructrs Amomi, Fructus Crataegi and Semen Raphani could promote the small intestinal propulsion significantly; Poria had little effect, and Glycyrrhiza, however, could inhibit small intestinal propulsion. As a result, the combination of Fructrs Amomi and massa medicata fermentata, Fructus Crataegi and Semen Raphani, Fructus Crataegi and Fructus aurantii immaturus could promote small intestinal propulsion by significant synergism.Conculsions1. The effect of Xiaoshiheweichongji on promotion of gastric emptying and small intestinal propulsion of normal mice fed with nutritional semi-solid food is similar to Mosapride, but better than Baohewan which can also improve gastrointestinal motility significantly.2. L—Arg can inhibit gastric emptying and small intestinal propulsion of mice significantly.3. Both Xiaoshiheweichongji and Mosapride can improve gastric emptying and small intestinal propulsion of mice'model, and the effects of the both have no difference to control group.4. seven component herbs of Xiaoshiheweichongji have different roles on promotion of gastric emptying and small intestinal propulsion. Fructrs Amomi, Fructus Crataegi and massa medicata fermentata can promote gastric emptying; Fructrs Amomi, Fructus Crataegi and Semen Raphani can promote small intestinal propulsion. Besides, the interaction of the component herbs also play a synergetic role on gastrointestinal motility.
