IntroductionDiabetes mellitus is a kind of chronic metabolic disease. The complications of diabetes mellitus have threatened patients more and more seriously, cerebral infarct is the most dangerous complication to diabetes mellitus patients.At present, the use of nerve protective agent and its therapeutic mechanisms to ischemic cerebrovascular diseases has become a focal point of research. Tongxinluo gelatin capsule is a kind of compound prepared traditional Chinese medicine, which was initiated by Professor Wu Yiling. It has invariable clinical therapeutic effect to ischemic cerebrovascular diseases.Insulin-like growth factor-1 (IGF-1) is a kind of neurotrophic factor, which has similar structure with insulin. It can regulate the growth and differentiation of neural cells, and play an important role in maintain the normal function of adult brain.The purpose of this study is to approach the protective mechanism of tongxinluo to diabetes mellitus complicating cerebral ischemia through detect the expression of IGF-1 in rats with diabetes mellitus complicating cerebral ischemia.Methods1,Experimental animals and groups:Male health Wistar rats (n=48) were made diabetes mellitus models and randomly devided into four groups: control group (n=12), sham operation group (n=12), MCAO group (n=12) and TXL group (n=12).2,To establish animal models: (1) Diabetes mellitus models: Wistar rats were intraperitoneal injected tetrao-xypyrimidine 200mg/kg and detected blood sugar 4 days later. The one whose fasting blood sugar > 11. 1mmol/L can be recognized diabetes mellitus rat.(2) MCAO models: Diabetes mellitus rats in MCAO group and TXL group subjected to 2-hour middle cerebral artery occlusion. Rats in sham operation group didn't subject middle cerebral artery occlusion.3,To detect the percentage of infarct volume:Six rats of each group were sacrificed and decapitated. TTC staining was performed to examine the percentage of infarct volume.4,To detect expression of IGF-1:Six rats of each group were sacrificed and decapitated to make paraffin section. Brain sections were analyzed by immunohistochemistry to determine the expression of IGF-1.5,Statistical Analysis:All data were expressed as x±s. The results were analyzed using t-test orANOVA and processed by SPSS 13.0. A value P<0.05 was considered statistically significant.Results1,The percentage of infarct volume:There were no infarct lesions in control and sham operation group. The percentage of infarct volume of MCAO group was significantly greater than that of TXL group (P<0.01) .2,Observation under microscope:There was no abnormality seen in control and sham operation group. There were ischemic changes in MCAO and TXL group.3,Detection of IGF-1: There were generous IGF-1 expression in MCAO and TXL group, which was significantly more than control and sham operation group(P<0.01), and the expression of IGF-1 in TXL group was significantly more than that in MCAO group.Conclusions1,TXL can decrease the infarction volume of rats with diabetes mellitus complicating cerebral ischemia;2,TXL can increase IGF-1 expression of rats with diabetes mellitus complicating cerebral ischemia;...
