| Castor Bean is the seed of Ricinus communis, which is belong to the familyEuphorbia. The main components of it are castor oil, ricin, ricinine ect. Theapplication in medicine of Castor Bean has a long history, such as in Compendium ofMateria Medica in Ming Dynasty, in Tang Materia Medica, ect.To roughtly separate the components of Castor Bean, we add 95% ethanol to thehomogenate of Castor Bean. The volume of the ethanol is about twice volume of thehomogenate. After stirring over night, the blend is centrifuged at 10,000rpm at 4℃for 30min. Then we can obtain two fractions, one fraction can dissovle in ethanol, theother can't. Remove the ethanol of the dissovled fraction to get a cream, thenrecirculate the cream with ether in a Soxhlet's apparatus for 12h to get the transparent oil that isthe Castor oil.The dregs is suspended in dilute HAc(PH=3.8) over night stirring. Then,centrifuge the blend to get the supernatant. To standardize the dregs, the proteincontent of supernatants was determined according to the method of Coomassie brilliantblue.Put the dregs in different conditions, investigate effects of defferent wet andtemperature on the anticontraception of ricin. The disposal of the dregs as follows:normal wet and temperature(control); 37℃, normal wet 1d; 37℃,high wet 1d;37℃,normal wet 2d;37℃,high wet 2d; 47℃,normal wet 1d;47℃,high wet1d;47℃, normal wet 2d;47℃, high wet 2d. The result demonstrate that the dregs showan obvious anticontraception on mice when put in normal wet and temperature. High temperature has a negtive effect on the anticontraception of the dregs. High humidityis extremely harmful to the preservation of Castor Bean's dregs.Simultaneously, we also take a view of the anticontraception of the castor oil.Whereas castor oil is liquid under normal temperature, we use complex coacerationto microcapsulate the castor oil. That is the liqid has been transferred to solid. Themicrocapsule have integrated coat when examined under light microscope and alsoshow a high ratio of oil-content. Animal experiment shows that the microcapsules ofcastor oil also have a good anticontraceptive effect. Additionally, to optimum theprocess of coacervation we adopt orthogonal design to the experiment. It shows thatwe can get the best mirocapsules when we emulsify the oil-water mixture at speed of1500rpm for 60min with the temperature at 60℃, PH at 4.0.Based on the above, we also inspect the anticontraceptive effect andsemi-lethal dosage of the mixture of the dregs and the microcapsules. The mixturenot only has a good anticontraceptive effect, but also shows great toxic effect onmice. |
