| Objective:The abuse of solvents has become a social problem in many countries. This research is to study the addiction of toluene of subchronic inhalation to rats and try to find the relationship between the olfactory pathway and the addiction of toluene of subchronic inhalation. To study the mechanism and the pathway of addictive materials of inhalation. To provide the empirical evidence of the prevention and cure of addictive materials of inhalation.Methods:Ninety rats were divided into nine groups: toluene concentrationâ… , concentrationâ…¡group, blank group, OB-impaired group, OB-impaired toluene concentrationâ… and concentrationâ…¡group, OE-injured group, OE-injured toluene concentrationand concentrationgroup. Rats were exposed to inhalation of concentrationâ… and concentrationâ…¡, or air only, in closed chambers for 4 weeks (5days/weed, 6 hours/day). Then, the addictions were examined. And the generating electricity of the OB in HAc group and toluene group were also examined by the system of Power Lab. The level of c-fos expression in NAc of control group, toluene concentration A and concentration B group were detected by immunohistochemistry after the detection of CPP. And the level of c-fos expression in OB(except the OB-impaired group, OB-impaired toluene concentration A and concentration B group) and NAc of each group without subchronic inhalation of toluene were detected by immunohistochemistry. Resultsâ‘ Compared to the blank group, the time in light chamber of toluene concentrationâ… and concentrationâ…¡group is longer (P<0.01),and in concentrationâ…¡group ,it is more longer(P<0.05). OB-impaired group, OB-impaired concentration and ? concentrationâ…¡group, OE-injured group, OE-injured concentrationâ… and ?concentrationâ…¡group show no CPP.â‘¡Electrophysiology: HAc group and electrophysiology toluene group show special generating electricity.â‘¢Compared to the control group, OB-impaired group and OE-injured group, OB-impaired toluene group and OE-injured toluene group, toluene induced significant c-fos expression in NAc of rats in other group. When detected the rats without subchronic inhalation of toluene, the c-fos expression in OB was significant in toluene concentrationâ… and concentrationâ…¡group. The results of c-fos expression in NAc were similar.ConclusionThe subchronic inhalation of solvent toluene can induce CPP of rats, which indicates that toluene is an addictive substance. And there may be new pathway of solvents addiction-the olfactory pathway. There may be a nervous pathway between OB or the senior olfactory center area and NAc. The pathway may be related to the mechanism of solvents addiction. |
