[Background]Ulcerative colitis(UC)is a chronic intestinal mucosa inflammatory disorder but its etiology has not been clearly understood.It is considered that its pathogenesis is associated with infection,heredity,immunity and oxidative stress et al,and oxidative stress might play a important factor in injury of mucosa in UC. Gastrointestinal mucosa is endowed with several antioxidant defense systems, including the reduced glutathione(GSH)focusing on neutralizing the damaging effects of ROS.Many studies had showed decreased levels of anti-oxidation agents including GSH.Theγ-GCS is the first enzyme involved and the rate determining step in glutathione biosynthesis,the expression ofγ-GCS in endochylema is association with GSH synthesis.Previous studies had showed the alteration of cytokines levels was correlated with pathogenesis of UC,it is known that the unbalance of Th1/Th2 might be a important factor in pathogenesis of UC, and immune system need be regulated by oxidative stress.The depletion of intracellular glutathione may influence the balance between Th1 and Th2 cells and have implications for allergic diseases.To study the expressionγ-GCS and Th1/Th2 in UC might help to identify the condition of oxidative stress and its association with disequilibrium of Th1/Th2.[objective]In this study,we explore the expression ofγ-GCS and the relationship betweenγ-GCS the expression of cytokines in colonic mucosa of UC,to study the role of and the expression ofγ-GCS and unbalance of Th1/Th2 in the pathogenesis and development of ulcerative colitis.[Methods] 1.The expression and role ofγ-GCS in colonic mucosa of ulcerative colitis patients1.1 Twenty eight biopsy specimen were taken from UC patients who had visited our hospital from August 2007 to February 2008.Ten colon polyp patients were controls.1.2 The expression ofγ-GCS in colonic mucosa were assayed in colonic mucosa of UC and normal controls by immunohistochemistry.2.The expression and role of IL-4,IFN-γand TNF-αin colonic mucosa of ulcerative colitis patients2.1 Twenty eight biopsy specimens were taken from UC patients who had visited our hospital from August 2007 to February 2008.Ten colon polyp patients were controls.2.2 Biopsy specimens were incubated in 5%CO2 and 7℃for 24h.2.3 The expression of IL-4,IFN-γ,TNF-αin colonic mucosa were assayed in culture supernatant of UC and normal controls.[Results]1.Theγ-GCS expressed in endochylema mainly,several positive cells expressed in mucosa of UC.The ratio ofγ-GCS in UC were less than those in controls. (P<0.01).2.The ratio of IL-4 in UC were less than those in control(0.1591±0.1014)(P<0.05),the ratio of IFN-γand TNF-αin UC(1.8349±1.6417)were more than in normal controls(1.4481±1.1661 1.8349±1.6417)(P<0.01).[Conclusion]1.The expression ofγ-GCS decreased significant in UC.2.The expression of IL-4 in UC were less than those in control,the expression of IFN-γand TNF-αin colonic mucosa is higher in UC than those in normal controls.The alteration of the three cytokines levels in intestinal mucosa might play the important role in the development of UC.3.The expression ofγ-GCS and disequilibrium of Th1/Th2 were detected in UC patients.It suggested that the correlation between oxidative stress and Th1/Th2.The mechanism is to be determined.
