Objective:To study if the effect that rhG-CSF suppresses the function of donor's CD4~+T cells and induces Th2 differentiation in PBSCT was associated with the impair of LFA- 1/ICAM- 1 co-stimulatory signaling.Methods:(1)CD4~+ T cells were separated by MACS from healthy donor's peripheral blood,before and after rhG-CSF was administrated.(2)Purified CD4~+T cells were stimulated by OKT3 and ICAM-1 which indue LFA-1/ ICAM-1 Signaling.(3)The marker of the activation of CD4~+ T cells such as CD69,CD25 and CD54 was detected by Flow cytometry after stimulated by OKT3 and ICAM-1.(4)The proliferation rate of CD4~+ T cells was detected by MTT after stimulated by OKT3 and ICAM-1.(5)The expression of IFN-γ,and IL-4 mRNA was evaluated by RT-PCR and realtime PCR analysis.Results:CD4~+ T cells were separated by MACS from healthy donor's peripheral blood,before and after rhG-CSF was administrated,and were stimulated by OKT3 and ICAM-1,(1)the expression of CD69,CD54 and CD25 after mobilization was significantly lower than the expression before mobilization(all P<0.01).(2)the proliferation rate after mobilization was significantly lower than that before mobilization.(3)the expression of IFN-γmRNA was decreased and IL-4 mRNA was increased after rhG-CSF was administrated.Conclusions:After rhG-CSF was administrated,the activation and proliferation of donor's CD4+T cells were suppressed and Th cells were induces to Th2 differentiation,which was associated with the impair of LFA-1/ICAM-1 signaling.
