Takingα-C octanoyl acetal sodium sulfite derivatives,viz.,methyl octanoyl acetal sodium sulfite,ethyl octanoyl acetal sodium and butyl octanoyl acetal sodium sulfite as tested sample,the effect of them on the production of acid phosphotase,lysozyme and SOD by macrophages in vivo were researched;a simple and effective method was set up for estimating the phagocytic function of macrophages in vivo,and the phagocytic function of macrophages being treated byα-C octanoyl acetal sodium sulfite derivatives was studied with it.α-C octanoyl acetal sodium sulfite derivatives can enhance macrophages to secrete acid phosphotase to different extend.The augmentation of methyl octanoyl acetal sodium sulfite and ethyl octanoyl acetal sodium was more notability than octanoyl acetal sodium sulfite(p<0.05).The augmentation of three derivatives depended on dosage.At a fixed derivative,the level of acid phosphotase flapped up linearly with the increase of concentration within the range from 0μg/ml to 120μg/m(p<0.01).The correlative coefficients of methyl octanoyl acetal sodium sulfite,ethyl octanoyl acetal sodium and butyl octanoyl acetal sodium sulfite are 0.970,0.958,0.937, respectively.The capability of enhancing macrophages to secrete acid phosphotase of methyl octanoyl acetal sodium sulfite was most obvious.But the capability was weakened when the number ofα-carbon atom exceeded a certain range(-C2H5),the capability of butyl octanoyl acetal sodium sulfite was the weakest.The lysozyme activity in macrophages stimulated by methyl octanoyl acetal sodium sulfite and ethyl octanoyl acetal sodium,behaved an extensively improvement trend(p<0.05).Not only the improvement trend of butyl octanoyl acetal sodium sulfite is not obvious,but also the level of lysozyme was decreased at the low concentration(15μg/ml).The research result indicated that methyl octanoyl acetal sodium sulfite and ethyl octanoyl acetal sodium might activate macrophages. The correlativity of concentration to the level of lysozyme was quite well(p<0.01).Within the concentration range from 0μg/ml to 60μg/ml,the correlative coefficients of methyl octanoyl acetal sodium sulfite,ethyl octanoyl acetal sodium and butyl octanoyl acetal sodium sulfite are 0.767, 0.861,0.785,respectively.At a fixed concentration,the level of lysozyme decreased linearly with the increase of the number ofα-carbon atom(p<0.0t).From 15μg/ml to 60μg/ml,the correlative coefficients are 0.867,0.882,0.738,respectively.The threeα-C octanoyl acetal sodium sulfite derivatives all can obviously enhance macrophages to secrete SOD(p<0.05).Compared with octanoyl acetal sodium sulfite,the capability of methyl octanoyl acetal sodium sulfite to stimulate macrophages to secrete SOD was the strongest (p<0.05)among the three derivatives;the capability of ethyl octanoyl acetal sodium was also improved further at medium and high concentration(30,60μg/ml).The correlativity of concentration to the level of SOD was quite well(p<0.01)within the concentration range from 15μg/ml to 60μg/ml.The correlative coefficients of methyl octanoyl acetal sodium sulfite,ethyl octanoyl acetal sodium sulfite and butyl octanoyl acetal sodium sulfite were 0.905,0.891,0.961, respectively.At a fixed concentration,the level of SOD decreased linearly with the increase of the number ofα-carbon atom(p<0.01).From 15μg/ml to 60μg/ml,the correlative coefficients are 0.783, 0.988,0.922,respectively.Taking octanoyl acetal sodium sulfite as tested sample,a method was set up for estimating the phagocytic function of macrophages in vivo.The method is easily handled,using the method can obtain true results,observe alive test phenomena and cost little.It could be considered as an effectively method for estimating the phagocytic function of macrophages in vivo.After being treated byα-C octanoyl acetal sodium sulfite derivatives,the bulk of macrophages was augmented obviously,the shape was abnormity and many pseudopods and prominencies protrude,which were the shape character of macrophages being activated.Compared with octanoyl acetal sodium sulfite,the threeα-C octanoyl acetal sodium sulfite derivatives all can obviously increase the phagocytosing percentages and phagocytosing indexes of macrophages of mice in vivo (p<0.05).The correlativity of concentration to the phagocytosing percentages and phagocytosing indexes of macrophages were quite well(p<0.01)within the concentration range from 25μg/ml to 50μg/ml.The correlative coefficients of methyl octanoyl acetal sodium sulfite,ethyl octanoyl acetal sodium sulfite and butyl octanoyl acetal sodium sulfite were0.968,0.999,0.930 and 0.932,0.929, 0.953,respectively.The capability of increasing the phagocytosing percentages and phagocytosing indexes of macrophages of methyl octanoyl acetal sodium sulfite was the strongest.But the capability was weakened when the number ofα-carbon atom reached to two,the capability of butyl octanoyl acetal sodium sulfite was the weakest.The correlativity of the number ofα-carbon atom to the phagocytosing percentages and phagocytosing indexes were quite well(p<0.01).With the concentration range from 15μg/ml to 60μg/ml,the correlative coefficients were were 0.943,0.932, 0.884 and 0.943,0.950,0.974,respectively.The result showed that theα-C octanoyl acetal sodium sulfite derivatives can obviously improve the phagocytosing function of macrophages(p<0.05), which could further prove that theα-C octanoyl acetal sodium sulfite derivatives have good effects on improving the phagocytosing function of macrophages and boost up the immunization of body.
