ObjectiveThrough anti-influenza A virus and IPF in vitro , screen of effective Chinese proprietary, named Huaxianyin; Apply Huaxianyin to treat IPF in clinic, Observe clinical efficacy, lay the foundation for the harmacological treatment of influenza A and IPF.Experiment Study1. Take the MDCK cell as the viral host, Determination of the Huaxianyin inhibit influenza A virus caused MDCK cells in lesions. And set Chinese medicine control group Kangbingdu oral liquid and western medicine control group Tamiflu. Observe the toxicity of MDCK cells of the Huaxianyin and Kangbingdu injection. Identify half of the toxic drug concentrations (TD₅₀) and the largest non-toxic concentrations (TD₀). Use Reed～Muench method to measure influenza A virus half suppression System concentrations (IC₅₀) of the Huaxianyin and Kangbingdu oral liquid, and measure Selection Index (SI= TD₅₀/IC₅₀). The Huaxianyin in 1:186 dilution reaches TD₅₀; in 1:324 dilution reaches TD5Oand IC₅₀, SI=4. 0. The Kangbingdu oral liquid in 1: 6.9 dilution reaches TD₅₀; in 1: 12 dilution reaches TD₀; in 1: 12 dilution reaches IC₅₀, SI=1. 7. The Tamiflu to the influenza virus A half inhibitory concentrationIC₅₀=22.2μg/ml. The results indicate : the Huaxianyin effects the cytopathicinduced by influenza A virus to a certain extent, than the Chinese medicine control group Kangbingdu oral liquid well, but the result is less than the western medicine control group.2. The 120 healthy bandicoots are randomly assigned to NGT and model set and Huaxianyin group and control group Prednisone Acetate,there are 30 of every group. Copy the IPF modle by the way of splash bleocin into trachea, Then kill the bandicoots in batches , one third of each group after accept 7 days' treatment, one third after 14 days, and after 28 days kill the remanent. At the same time, observe the change of pathplogy, collagen contents,cell count. The results indicate :Huaxianyin has the effect of against IPF, in the way of improving pathplogy change,reducing collagen contents and cell count,and keep down the form of IPF. Clinic StudyObserve clinical of the Huaxianyin treating IPF in clinic. Compliance with the standards included in the positive entering the study, and select 16 patients. 16 patients are randomly assigned to the Huaxianyin group and control group Prednisone Acetate, the differences of two groups in terms of gender, age, severity level are not significant, statistical comparability (P>0.05).Use the Huaxianyin to treat IPF, and the total efficiency is 66.66%,the control group Prednisone Acetate is 57.14%. There hasn't statistical comparability (P>0.05).Clinical results suggest:the Huaxianyin has the same effect with Prednisone Acetate to IPF.SummaryExperiments in vitro show that, the Huaxianyin can inhibit cell disease caused by influenza A virus, it is better than the Chinese medicine control group Kangbingdu oral liquid, But it is not as great as the Western medicine Tamiflu.In vivo observation,Huaxianyin has the effect of against IPF.In clinical observation, although Huaxianyin isn't better than control group, It is still suggested that the role of Chinese medicine against IPF might be the same as Prednisone Acetate.
