| Objective:(1)Study on intestinal microflora in children with Henoch-Schonlein purpura(HSP)before and after treatment.(2)To guide probiotics in the preparation of Henoch-Schonlein Purpura treatment.Methods:(1)The feces of 43 healthy children(control group)and 43 children with HSP(observation group)1st day before as well as third and seventh day after the therapy were collected.The bacterial DNA in the feces were extracted,bacterial DNA-OD260was measured by biophotometer.(2)16SrRNA/DNA fluorescent quantitative PCR were applied in compare the bacterial content of Bifidobacterium,Lactobacillus and Escherichia coli in feces among control group and different observation group.Results:(1)Bacteria DNA-OD260in the feces is respectively:control group d03605.9±1096.9ng/μl,observation group 1st day before treatment d12225.9±616.1ng/μl,the 3th day d21780.3±547.4ng/μl,and the 7th day d32055.6±570.2ng/μl;Compared control group and the observation group,d0 and d1 have difference(P<0.05),d1 and d2 have differences(P<0.05),d1 and d3 have differences(P<0.05),d2 and d3 no differences(P>0.05);(2)The Bifidobacterium logarithm in the feces is respectively:control group,9.47±0.41,observation group 1st day before treatment 7.44±0.87,the 3th day after treatment 5.97±0.81,the 7th day after treatment 6.67±0.87;The Lactobacillus logarithm in the feces is respectively:control group 9.18±0.48,observation group the 1st day before treatment 6.52±0.64,the 3th day after treatment 5.27±0.77,the 7th day after treatment 5.93±0.65;The Escherichia coli logarithm in the feces is respectively:control group,7.55±0.41,observation group the 1st day before treatment 6.91±1.25,the 3th days after treatment 5.93±1.08,the 7th day after treatment 6.41±1.12;The amount of Bifidobacterium and Lactobacillus in the feces among healthy group,pre-therapy group and post-therapy group are significantly different(P<0.05),especially on the 3th day,which begin to rise on the 7th day;E.coli in feces samples of some of the numerical reduction,the 3th day of the lower,but the 7th day did not rebound.Conclusions:(1)The amount of total bacteria in the feces between healthy children and 1st day before treatment in children with HSP have significant differences. (2)Compared with healthy children,the amount of the intestinal Bifidobacterium, Lactobacillus and E.coli in children with HSP is decrease.(3)The decrease of the intestinal flora may be influence on the outbreak and prognosis of the HSP. |
