As a kind of chemokines, stromal cell derived factor-1α(SDF-1α) and its receptor CXCR4 play an important role in embryogenesis, tumor metastasis and mediation HIV infection. Latest research found that SDF-1α/CXCR4 was highly expressed in atherosclerosis plaque. And it is closely related with inflammation of vessel wall. So its role in atherosclerosis is very important.Firstly, it was explored in our study that the effect of SDF-1α/CXCR4 on THP-1 chemotaxis, THP-1/ECV304 cells adhesion and the effect of advanced oxidation protein products (AOPP) to chemotaxis and adhesion. Then SDF-1αexpression were research in ECV304 cells pretreated with AOPP.Objective To investigate the effect of SDF-1αon THP-1 cells chemotaxis and influence of AOPP, the effect of SDF-1α/CXCR4 on THP-1/ECV304 cells adhesion, and the effect of AOPP on the expression of SDF-1αin ECV304 cells.Methods THP-1 cells chemotaxis was examined with transwell permeable supports. THP-1 cells with or without pretreated with AOPP were input in upper chambers, SDF-1αwas added in low chambers. Number of migrated cells in low chamber was counted after incubation for 10 hours.ECV304 cells pretreated with 50μmol/l AOPP for 24 hours were incubated with THP-1 cells to observe cells adhesion. SDF-1αmRNA was revealed by RT-PCR respectively in ECV304 cells incubated with different concentrations of AOPP.Results1. SDF-1αinduced a pronounced migration of input cells in a concentration-related manner, but this effect was obviously inhibited by the antibody to CXCR4. After pretreated with different concentration of AOPP for 24 hours, THP-1 cells were exposed to 10 ng/ml SDF-1α, THP-1 cells migration enhanced by AOPP in a concentration-dependent manner was found. 2. THP-1/ECV304 cells adhesion was hanced by AOPP in a concentration- dependent manner.3. SDF-1αwas constitutionally expressed in ECV304 cells and was up-regulated by AOPP in a concentration-dependent manner.Conclusions1. SDF-1α/CXCR4 is implicated in THP-1 cells chemotaxis and chemotactic response of monocyte to SDF-1αis enhanced by AOPP.2. SDF-1α/CXCR4 is implicated in THP-1/ECV304 cells adhesion and its adherent activity is enhanced by AOPP.3. AOPP up-regulate SDF-1αexpression.
