The First Step Studies To Ameliorate L-J Culture Medium Further

Background: L-J medium that takes egg as basis is one of solid culture medium of training Mycobacterium tuberculosis and always apply to clinical broadly,it is one of most commonly used culture medium as training Mycobacterium tuberculosis at present. Whose characteristic is that moderate price, the manufacture method is simple, easy to observe colony shape,etc. There have not read that news report of general growth bacterium on L-J culture medium since the culture medium has some malachite green now(the mould does not include). During the working period,the author observes one streptococcus(S) on L-J culture medium and preserves it to study. Objective: To study the growth of the streptococcus and Mycobacterium tuberculosis on old and new L-J culture mediums and to ameliorate L-J culture medium further.Study 1 To add a little different bacteriostat and to observe growth of the streptococcus and the tuberculosis on two kinds of L-J culture mediumMethods: The streptococcus and one M.TB and one M.bovis from the clinical and one H37Rv as QC tuberculosis,the 4 Bacterium in total is inoculated on L-J culture mediums that add a little Carbenicillin(50.0mg/ml)and/or NaSeO4(5,10,20μg/ml), the disinoculated medium as blank.Culture these 4 weeks to observe on 37±0.2℃.Results: The new L-J culture medium that has a little Carbenicillin and NaSeO4 holds growth of the streptococcus back in different extent.Which have 20μg/ml NaSeO4,or Carbenicillin and 10μg/ml NaSeO4,or Carbenicillin and 20μg/ml NaSeO4 always restain growth of the streptococcus completely. Ones which contain 20μg/ml NaSeO4,Carbenicillin and 20μg/ml NaSeO4 restain slightly,the others has no obvious to hold growth the 3 TBs back. Study 2 Observe to the growth condition of the random 30 TBs inoculated on old and new improving L-J culture mediumsMethods: The random 30 TBs that our hospital separate from clinical during the past almost 3 months are inoculated on two L-J culture medium.Observed to the growth condition when cultured 4 weeks on 37±0.2℃.Result: The 30 TBs on two kinds of culture mediums are all being, but it is different in their areas of growth. Maked use of statistics software SPSS 10.0 to check (P=0.504/0.767/0.748) and confirmed to have little notable difference.Study 3 Observe and analyze to the 384 clinical samples which were inoculated on both the old and the new L-J acid culture mediumsMethods: The 384 samples which we collected the types of specimens from January 1, 2008 to January 31,were inoculated on both the old and the new L-J acid culture mediums.Results were observed for four weeks on 37±0.2℃. Result: After four weeks,the 384 clinical specimens were cultured on 37±0.2℃and the 98 cases confirmed positive.There are 45 M.TBs,32M.Bs and 16 MOTTs on the OLD L-J acid medium;there are 49 M.TBs,33M.Bs and 16 MOTTs on the NEW L-J acid medium improvement on the acid medium.But nine cases of the OLD L-J acid medium are polluted, and four cases of the NEW L-J acid medium are polluted.Theχ2 test shows that two kinds of L-J acid mediums have no significant difference.Conclusion: Specimen nitrification(4% NaOH or 4% H2SO4 handles) and malachite green added by L-J culture medium inner can not restrain the general bacterium's completely growth, whose cultivates still having a little disturbance to TB.It needs to reform L-J culture medium further therefore. What be added Carbenicillin (50.0mg/ml) and NaSeO4 (10mg/ml) has no obvious effect on the new L-J culture medium to the growth of TB.And the average bacterium has certain inhibitory action.As a result, the further experiment studies looks on that component as the new ameliorating L-J culture medium.
